摘  要:花粉发育和花粉管生长过程与SNAP30介导的囊泡运输密切相关,前期结果暗示SNAP30基因可能正向调控了马铃薯品种‘青薯9号’的花粉发育和自交结实过程。为了进一步研究囊泡运输相关蛋白StSNAP30在马铃薯花粉发育和花粉管生长中的生物学功能。本研究以‘青薯9号’花器官为材料,对StSNAP30进行了克隆,并进行了蛋白理化性质、系统进化、表达模式、亚细胞定位和启动子序列分析。结果显示,StSNAP30基因CDS序列全长798 bp,编码了265个氨基酸,蛋白分子量约29 kD,理论pI为6.63。StSNAP30蛋白包含Qb-SNARE和Qc-SNARE结构域,不具有跨膜结构域。StSNAP30与双子叶植物SNAP30蛋白聚为一类,该蛋白亚细胞定位于液泡膜和细胞膜。StSNAP30特异性地在花器官中表达,基因表达水平随着花粉发育进程而不断升高。StSNAP30启动子中具有多个环境响应调控元件。本研究结果表明StSNAP30蛋白是一个典型的Qbc-SNARE蛋白,可能介导液泡和质膜间囊泡转运过程,参与了马铃薯成熟花粉的萌发和花粉管生长过程。本研究为进一步解析StSNAP30对花粉发育的调控功能奠定了理论基础。The pollen development process and pollen tube growth are closely related to vesicle transportation mediated by SNAP30.Previous work suggested SNAP30 might positively regulate the pollen development process and self-fruitfulness of potato cultivar'Qingshu No.9'.In order to explore the biological function of StSNAP30,the StSNAP30 codon sequence was cloned from'Qingshu No.9'flower materials,and the physical and chemical properties of proteins,systematic progression,expression patterns,subcellular localization and promoter sequence analysis were carried out in this study.The results showed that the StSNAP30 CDS sequence is 798 bp,encoded 265 amino acids,the protein molecule is about 29 kD,and the theoretical pI is 6.63.StSNAP30 protein contains Qb-SNARE and Qc-SNARE domains,which do not have transmembrane domains.StSNAP30 is clustered with dicotyledons SNAP30 proteins.StSNAP30 was specifically expressed in flower organs,especially the expression level was increased with pollen development process.StSNAP30 protein ware subcellular localization to vacuoles and cell membranes.The promoter of StSNAP30 contains several environmental responses cis-elements.In summary,StSNAP30 protein is a typical Qbc-SNARE protein,probably participated in vesicular trafficking of vacuolar membrane and plasma membrane,and which may be involved in the germination and tube growth of potato mature pollen.The results of this study lay the foundation for further analysis of StSNAP30 biological function in potato pollen development process.
