摘 要:目的:探讨METTL3在乳腺癌组织中的表达及临床意义。方法:免疫组织化学检测乳腺癌及乳腺增生组织中METTL3的表达,分析乳腺癌组织中METTL3表达与乳腺癌临床病理因素的关系。siRNA敲除乳腺癌细胞MDA-MB231中的METTL3,观察对细胞增殖、迁移、侵袭的影响。结果:高表达METTL3在乳腺增生组织及乳腺癌组织中分别为25.0%和57.8%(χ^(2)=7.042,P=0.008);高表达METTL3在淋巴结阳性及淋巴结阴性中分别为68.3%和45.9%(χ^(2)=3.979,P=0.046);MTT细胞增殖实验提示METTL3沉默组细胞增殖能力降低,而划痕实验提示siRNA敲除METTL3明显抑制乳腺癌细胞MDA-MB231的迁移(P<0.05)。细胞侵袭实验提示si-METTL3抑制了乳腺癌细胞MDA-MB231的侵袭(P<0.05)。结论:乳腺癌组织中METTL3表达增高,高表达METTL3与乳腺癌淋巴结转移有关。敲除METTL3抑制乳腺癌MCF-7细胞增殖、迁移及侵袭。乳腺癌中METTL3高表达与恶性程度有关,METTL3可能是乳腺癌治疗的一个新靶点。Objective:To investigate the expression and mechanism of METTL3 on breast cancer.Methods:METTL3 expression in breast cancer tissues was detected by immunohistochemistry,METTL3 was knockdowned by siRNA,and the effects on MDA-MB231 cells proliferation,migration and invasion were observed.Results:The odds of METTL3 expression was 25.0%and 57.8%in breast hyperplasia tissues and breast cancer tissues,repectively(χ^(2)=7.042,P=0.008).1.The odds of METTL3 expression was 68.3%and45.9%in lymph node positive breast cancer tissues and ymph node negative breast cancer tissues,repectively(χ^(2)=3.979,P=0.046).MTT cell proliferation assay showed that cell proliferation was decreased in MettL3 silenced group,while scratch assay showed that siRNA knockout of MettL3 significantly inhibited migration in MDA-MB231 breast cancer cells.Cell invasion assay suggested that si-mettl3 inhibited the invasion of MDA-MB231 in breast cancer cells.Conclusions:METTL3 expression was increased in breast cancer tissues,high METTL3 expression was associated with lymph node metastasis of breast cancer.METTL3 might be involved in the progression of breast cancer,and the METTL3 might be a new therapeutic target.
