摘 要:目的:采用核磁共振氢谱(^(1)H NMR)技术研究加味黄连汤对应激性胃溃疡小鼠胃黏膜组织代谢物表达的影响。方法:40只SPF级KM小鼠随机分为正常组、模型组、西药组和中药组,每组10只。利用束缚-冷应激法制备应激性胃溃疡模型,造模成功后连续7 d分别用加味黄连汤中药汤液和艾司奥美拉唑镁肠溶片水溶液对中药组和西药组小鼠进行干预。采用HE染色法观察小鼠胃黏膜组织病理形态,采用;H NMR技术进行代谢组学检测,通过模式识别方法分析各组间的代谢轮廓差异,并筛选出差异性代谢物,通过在线分析网站寻找相关代谢通路。结果:正常组小鼠胃黏膜上皮结构完整,腺体紧密,细胞排列整齐;模型组小鼠胃黏膜上皮结构缺损,腺体散乱,细胞排列紊乱,可见炎症细胞浸润和毛细血管出血;中药组和西药组小鼠胃黏膜上皮结构完整,无充血及水肿。胃黏膜组织代谢组学共筛选出有关代谢标志物13种,参与了15条代谢通路。结论:加味黄连汤能够调节胃溃疡模型小鼠胃黏膜组织代谢物的表达,促进胃溃疡愈合。Objective: To investigate the effect of Modified Huanglian Decoction on the expression of gastric metabolites in mice with stress gastric ulcer by ^(1)H NMR. Methods: Forty SPF KM mice were randomly divided into normal group, model group, western medicine group and traditional Chinese medicine group, with 10 mice in each group. Mice model of stress gastric ulcer were prepared by the method of restraint-cold stress. After successful modeling, mice in the western medicine group and the traditional Chinese medicine group were gavaged with esomeprazole magnesium enteric-coated tablet aqueous solution and Modified Huanglian Decoction for seven days. Histopathological changes of the gastric tissue were observed by HE. Analyzing the metabolic profile of differences between groups by pattern recognition methods, selecting different metabolites, finding related metabolic pathways through the analysis of the online website. Results: The gastric mucosa epithelium of normal mice was intact,and the glands were compact and cells were arranged neatly. In the model group, gastric mucosal epithelium was defective,glands were cluttered and cells were disordered, inflammatory cell infiltration and capillary bleeding were observed. The epithelial structure of gastric mucosa of mice in TCM group and western medicine group was intact without hyperemia and edema. A total of 13 metabolic markers were identified by gastric tissue metabonomics, participating in 15 metabolic pathways. Conclusion:Modified Huanglian Decoction can regulate the expression of metabolites in gastric mucosa and promote gastric ulcer healing in gastric ulcer model mice.
