摘 要:以枇杷叶为原料,研究微波辅助提取熊果酸和齐墩果酸的最优工艺。采用高效液相色谱法测定熊果酸和齐墩果酸含量,以熊果酸和齐墩果酸的总得率为评价指标,在单因素试验的基础上,采用Box-Behnken设计-响应面法优化提取工艺。结果表明,熊果酸和齐墩果酸的分离度为2.67,浓度分别在0.2~1.2mg/mL、0.02~0.12mg/mL范围内与峰面积呈良好的线性关系;最优提取工艺为提取时间11.5min、提取温度75℃、料液比1∶20g/mL、乙醇体积分数100%、微波功率500W,在此工艺条件下熊果酸和齐墩果酸的总得率为0.928%,与模型预测值相对误差为1.25%。优化的提取工艺条件稳定、可靠,适用于枇杷叶中的熊果酸和齐墩果酸的提取。The optimized microwave-assisted extraction technology of ursolic acid(ursolic acid,UA)and oleanolic acid(oleanolic acid,OA)from eriobotryae folium was studied.HPLC was used to determine the content of UA and OA.Box-Behnken design-response surface method was conducted to optimize the extraction process based on the single factor tests,when the total yield of UA and OA was used as evaluated index.The results showed that the concentrations of UA and OA showed good linear relationships in the ranges of 0.2~1.4 and 0.02~0.14mg/mL,respectively,with a resolution of 2.67;the optimal extraction process was as follow:extraction time was 11.5min,extraction temperature was 75℃,the ratio of material to liquid was 1∶20,and the concentration of ethanol was 100%.Under this condition,the total yield of UA and OA was 0.928%,which was 1.25% to the relative error from the theoretical prediction.The optimal extraction technology was stable and reliable,which would be suitable for the extraction of UA and OA from eriobotryae folium.
