摘 要:目的:制备薰衣草精油纳米乳,考察其理化性质及体外抑菌活性。方法:采用相转变法制备薰衣草精油纳米乳,通过伪三相图法优化薰衣草精油纳米乳最优配方。通过粒径分析仪、透射电镜测定其粒径分布和形态。测定薰衣草精油纳米乳对金黄色葡萄球菌和大肠杆菌的体外抑菌活性。结果:薰衣草精油纳米乳最优配方为20.91%氢化蓖麻油、6.97%聚乙二醇-400、6.97%薰衣草精油、65.15%蒸馏水。此条件下制备的薰衣草精油纳米乳澄清、透明、流动性好,平均粒径为23.79±0.30 nm,分散指数为0.443±0.12,呈正态分布;透射电镜下其形态为球形,分布均匀、分散性良好,载药量为53.95±1.12 mg/mL。体外抑菌实验表明,薰衣草精油纳米乳对金黄色葡萄球菌和大肠杆菌的抑菌圈直径分别为13.67±0.33 mm和11.87±0.13 mm,而薰衣草精油的抑菌圈直径分别为9.75±0.20 mm和8.93±0.16 mm,薰衣草精油纳米乳抑菌效果优于薰衣草精油。结论:试验制备的薰衣草精油纳米乳制备工艺简单,体外抑菌效果明显,值得进一步研究。Objective:To prepare lavender essential oil nanoemulsion and to investigate its physical and chemical properties and antibacterial activity in vitro. Methods:The lavender essential oil nanoemulsion was prepared by the phase inversion method,and the optimal formula of the lavender essential oil nanoemulsion was optimized by the pseudo three-phase diagram method. The particle size distribution and morphology were measured by particle size analyzer and transmission electron microscope. In vitro antibacterial activity of lavender essential oil nanoemulsion against Staphylococcus aureus and Escherichia coli was determined. Results:The optimal formula of lavender essential oil nanoemulsion was 20.91% hydrogenated castor oil,6.97% polyethylene glycol-400,6.97% lavender essential oil,and 65.15% distilled water. The lavender essential oil nano emulsion prepared under this condition was clear,transparent and has good fluidity. The average particle size was 23.79 ± 0.30 nm and the dispersion index was0.443 ± 0.12;Under transmission electron microscope,it was spherical,evenly distributed and well dispersed,and the drug loading was 53.95 ± 1.12 mg/mL. In vitro bacteriostatic experiments showed that the bacteriostatic circle diameters of lavender essential oil nanoemulsion against Staphylococcus aureus and Escherichia coli were 13.67 ± 0.33 mm and 11.87 ± 0.13 mm respectively,while the bacteriostatic circle diameters of lavender essential oil were 9.75 ± 0.20 mm and 8.93 ± 0.16 mm respectively. The bacteriostatic effect of lavender essential oil nanoemulsion was better than that of lavender essential oil.Conclusion:The lavender essential oil nanoemulsion prepared in this experiment has a simple preparation process and obvious antibacterial effect in vitro,which is worthy of further study.
