摘  要:目的:为确定超声辅助提取鸡桑叶总黄酮的最佳工艺并考察总黄酮的抗氧化活性。方法:首先考察乙醇浓度、液料比、超声温度和超声时间4个单因素对鸡桑叶总黄酮提取率的影响,再通过Box-Behnken方法对影响鸡桑叶总黄酮提取率的工艺因素进行响应面优化,最后通过鸡桑叶总黄酮对DPPH·和·OH清除效果来评价其抗氧化活性。结果最佳的工艺条件为乙醇浓度73%,液料比21 mL/g,超声温度63℃和超声时间25 min,得到鸡桑叶总黄酮的提取率为37.08 mg/g,与理论值的相对误差为1.72%。鸡桑叶总黄酮具有一定的清除DPPH·和·OH的能力,并呈一定的量效关系,其IC50分别为60.51 mg/L和156.05 mg/L。结论:利用响应面对鸡桑叶总黄酮的超声辅助提取工艺进行优化,拟合度好,误差小,准确率高,所得的鸡桑叶总黄酮具有一定的抗氧化能力,是一种潜在的天然抗氧化剂。Objective:In order to determine the best technology of ultrasonic-assisted extraction and its antioxidant activity of total flavonoids from leaves of Morus australis.Methods:Firstly,the influence of four single factors of ethanol concentration,liquid-to-material ratio,ultrasonic temperature,and ultrasonic time on the extraction yield of total flavonoids from leaves of M. australis was investigated. Secondly,the technological factors affecting the extraction yield of total flavonoids from leaves of M. australis was optimized by Box-Behnken method. Finally the antioxidant activity was evaluated through the scavenging effect of total flavonoids from leaves of M. australis on DPPH· and ·OH.Results:The optimum extraction conditions were ethanol concentration 73%,liquid to material ratio 21 mL/g,ultrasonic temperature 63℃,ultrasonic time 25 min. The yield of actual total flavonoids was 37.08 mg/g under the optimal extraction condition with the relative error of 1.72% compared to the predictive value. The total flavonoids from leaves of M. australis had a certain ability to clear DPPH· and ·OH,and showed a certain dose-effect relationship,with IC50of 60.51 mg/L and 156.05 mg/L,respectively.Conclusion:The response surface methodology was used to optimize the ultrasonic-assisted extraction process of total flavonoids from leaves of M. australis,with a good fit,small error,and high accuracy. The obtained total flavonoids had a certain antioxidant capacity and would be a potential natural antioxidant.
