摘  要:目的研究脑蛋白水解物-Ⅰ(CH-Ⅰ)对帕金森病(PD)小鼠的神经保护作用及其机制。方法成年健康雄性C57BL/6小鼠48只,随机分为对照组、模型组、CH-Ⅰ10 mg/kg组和CH-Ⅰ20 mg/kg组。腹腔注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)诱导PD小鼠模型,腹腔注射CH-Ⅰ进行干预治疗。通过悬挂实验评价各组小鼠的神经行为功能,采用Nissl染色观察纹状体神经细胞的形态结构,采用免疫印迹法检测纹状体神经细胞内酪氨酸羟化酶(TH)、脑源性神经营养因子(BDNF)和神经生长因子(NGF)的表达。结果模型组小鼠的机体协调性较对照组明显减弱,而CH-Ⅰ10 mg/kg组和CH-Ⅰ20 mg/kg组小鼠的机体协调性则较模型组明显改善(F=5.516,P<0.05)。与模型组相比,CH-Ⅰ10 mg/kg组和CH-Ⅰ20 mg/kg组小鼠纹状体神经细胞NGF、BDNF和TH的表达明显增加(F=26.567~43.458,P<0.01)。Nissl染色显示,CH-Ⅰ干预可有效减轻MPTP诱导的细胞损伤。结论CH-Ⅰ可通过调节神经营养因子和TH的表达水平对PD小鼠发挥神经保护作用。Objective To investigate the neuroprotective effect and mechanism of cerebroprotein hydrolysate-Ⅰ(CH-Ⅰ)on mice with Parkinson’s disease(PD).Methods A total of 48 healthy adult male C57 BL/6 mice were randomly divided into control group,model group,CH-Ⅰ10 mg/kg group,and CH-Ⅰ20 mg/kg group.A mouse model of PD was established by intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP),and intraperitoneal injection of CH-Ⅰwas given as intervention treatment.The wire hanging test was used to evaluate neurobehavioral function;Nissl staining was used to observe the morphology and structure of neural cells in the corpus striatum;Western blotting was used to measure the expression of tyrosine hydroxylase(TH),brain-derived neurotrophic factor(BDNF),and nerve growth factor(NGF)in neural cells of the corpus striatum.Results Compared with the control group,the model group had a significant reduction in body coordination,and compared with the model group,the CH-Ⅰ10 mg/kg group and the CH-Ⅰ20 mg/kg group had a significant improvement in body coordination(F=5.516,P<0.05).Compared with the model group,the CH-Ⅰ10 mg/kg group and the CH-Ⅰ20 mg/kg group had significant increases in the expression of NGF,BDNF,and TH in neural cells of the corpus striatum(F=26.567-43.458,P<0.01).Nissl staining showed that CH-Ⅰintervention effectively alleviated cell injury induced by MPTP.ConclusionCH-Ⅰmay exert a neuroprotective effect on PD mice by regulating the expression of neurotrophic factors and TH.
