摘 要:厌氧氨氧化细菌生长缓慢,颗粒化有助于其在反应器中的持留,防止污泥流失,而胞外多糖作为颗粒化的生物胶,是其颗粒化的重要因素,因此研究其胞外多糖的代谢途径及结构对厌氧氨氧化工艺的推广应用至关重要。但是,目前尽管厌氧氨氧化污泥的胞外聚合物的提取方法众多,但尚无针对其胞外多糖提取方法的比较研究。因此,该研究对比了3种胞外聚合物(EPS)的提取方法:甲酰胺-NaOH法、加热-Na_(2)CO_(3)法和甲醛-NaOH法。通过比较其污泥形态、EPS产量、TOC总量、3D-EEM图谱及污泥细胞裂解程度,发现甲醛-NaOH法所得到的EPS提取液胞外多糖含量最高、胞外蛋白含量最低、细胞裂解程度最低,是厌氧氨氧化颗粒污泥中胞外多糖的最佳提取方法。该研究可为厌氧氨氧化污泥胞外多糖的代谢途径和结构研究提供技术支撑。Anammox bacteria have long been known as its slow-growing.Granulation is benefit to prevent sludge loss by retaining microorganisms within the reactor,and polysaccharide as the biological gels determined the degree of granulation.So that,it is essential to explore the metabolic pathways and molecular structures of polysaccharides.Although there are many methods for the extraction of extracellular polymeric substances(EPS)from Anammox bacteria,but there still little reports about the comparative studies on the extraction of extracellular polysaccharides.Here we compared three EPS extraction methods,formamide with NaOH,heating-Na_(2)CO_(3)and formaldehyde with NaOH,and found that EPS obtained by formaldehyde with NaOH had the highest polysaccharide content,the lowest protein content and the lowest degree of cell lysis,by comparing the morphological characteristics of sludge,EPS yields,TOC contents,3D-EEMs fluorescence spectra and the degree of cell apoptosis.Therefore,formaldehyde with NaOH is the best approach to obtain the extracellular polysaccharides from Anammox bacteria and the study is helpful to understanding the metabolic pathways and molecular structures of extracellular polysaccharides.
