摘　　要：目的:基于“药辅合一”的理念,优化桉叶油乳膏的制备工艺,并对其进行药效学研究。方法:以桉叶油、硬脂酸和单硬脂酸甘油酯的用量为考察因素,采用Box-Behnken响应面法优化桉叶油乳膏的制备工艺;通过小鼠耳肿胀实验和热板实验,分别考察桉叶油乳膏的抗炎作用和对小鼠痛阈值的影响;通过体外抑菌实验,考察桉叶油乳膏对金黄色葡萄球菌和大肠埃希氏菌的抑制作用。结果:桉叶油乳膏的最优基质处方为桉叶油用量1.1 g、硬脂酸用量1.0 g、单硬脂酸甘油酯用量1.6 g;制得的乳膏能显著抑制二甲苯所致小鼠耳廓肿胀和热板仪所致小鼠疼痛(P<0.05,P<0.01);制备的桉叶油乳膏对金黄色葡萄球菌和大肠埃希氏菌均表现出抑制作用,且对金黄色葡萄球菌的抑制能力优于大肠埃希氏菌。结论:优化后制备的桉叶油乳膏工艺简单可行,稳定性良好,有良好的抗炎、镇痛和抑菌作用,可为桉叶油的开发提供参考。Objective:To optimize the preparation technology of eucalyptus oil cream based on the concept of"unification of medicine and adjuvant"and investigate its pharmacologic effects.Methods:The preparation technology of eucalyptus oil cream was optimized by the Box-Behnken Response Surface Methodology with eucalyptus oil,stearic acid and glyceryl monostearate as factors.The anti-inflammatory effect of eucalyptus oil cream was observed through the mouse ear swelling test.The effect of eucalyptus oil cream on the pain threshold of mice was investigated by the hot plate test.The inhibitory effects of eucalyptus oil cream on Escherichia coli and Staphylococcus aureus were detected via the in vitro antibacterial assay.Results:The optimal prescription of eucalyptus oil cream consisted of 1.1 g of eucalyptus oil,1.0 g of stearic acid,and 1.6 g of glyceryl monostearate.The prepared eucalyptus oil cream could significantly suppress the xyleneinduced ear swelling of mice and relieve the pain caused by the hot plate(P<0.05,P<0.01).Eucalyptus oil cream showed inhibitory effects on both E.coli and S.aureus,and the inhibitory activity against S.aureus was stronger than that against E.coli.Conclusion:The optimized eucalyptus oil cream is simple in preparation and stable in property and shows good antiinflammatory,antibacterial,and analgesic effects.The present study is expected to provide a theoretical basis for the development of eucalyptus oil.
