摘  要:目的:研究穗花杉双黄酮(AF)对骨肉瘤细胞增殖的作用及其分子机制。方法:CCK-8法筛选AF抑制骨肉瘤细胞增殖的有效浓度,细胞周期流式法检测AF对骨肉瘤细胞G_(1)期和S期分布的影响,免疫荧光法、荧光定量PCR法和Western blot法检测AF对骨肉瘤细胞增殖相关基因及蛋白表达的影响。结果:AF在浓度为75μmol/L时,显著降低骨肉瘤细胞活力(P<0.05);促进骨肉瘤细胞停滞在G_(1)期,无法进入S期;抑制骨肉瘤细胞β-catenin核表达;AF上调p21 mRNA和蛋白表达(P<0.05),下调骨肉瘤细胞β-catenin和Cyclin D1 mRNA和蛋白表达(P<0.05)。结论:AF对骨肉瘤细胞增殖有抑制作用,其机制是上调p21、下调β-catenin和Cyclin D1的表达。Objective:To study the effect of amentoflame(AF)on the proliferation of osteosarcoma cells and its molecular mechanism.Methods:The effective concentration of AF to inhibit the proliferation of osteosarcoma cells was screened by CCK-8 method.The effect of AF on G_(1) and S phase distribution of osteosarcoma cells was detected by cell cycle flow cytometry.The effect of AF on proliferation-related genes and proteins expression of osteosarcoma cells was detected by immunofluorescence,fluorescence quantitative PCR and Western blot.Results:At a concentration of 75μmol/L,AF significantly reduced the viability of osteosarcoma cells(P<0.05);promoted the stagnation of osteosarcoma cells in the G_(1) phase and could not enter the S phase;inhibited the nuclear expression ofβ-catenin in osteosarcoma cells;upregulated p21 mRNA and protein expression down-regulatedβ-catenin and Cyclin D1 mRNA and protein expression in osteosarcoma cells(all P<0.05).Conclusion:AF inhibits the proliferation of osteosarcoma cells mainly by increasing the expression of p21 and decreasing the expression ofβ-catenin and Cyclin D1.
