摘  要:为深入了解鸡NOD样受体(NLRs)家族基因特性,利用生物信息学方法通过保守结构域搜索鉴定了鸡的NLRs基因,分析NLRs基因理化性质、基因结构、系统进化关系、正选择作用、共线性关系和表达情况。结果显示:共鉴定出5个鸡NLRs基因,分别命名为chNLRC5、chNLRC3、chNLRP3、chNLRX1、chNOD1,分布在5条染色体上,编码734~1 862个氨基酸,编码蛋白相对分子质量为82 842.01~207 969.52 ku,等电点为5.48~8.64;chNLRC5、chNOD1和chNLRP3为酸性蛋白,chNLRC3和chNLRX1蛋白呈碱性,只有chNOD1为稳定蛋白,5个成员均为亲水蛋白,5个基因结构差别显著,分别含有48、20、7、7、11个外显子;系统进化树显示,NLRs基因家族分为5个分支,鸡、绿头鸭、火鸡和日本鹌鹑NLR基因间存在较强的共线性关系;对各成员进行选择压力分析结果鉴定出13个正选择位点;鸡回肠和肺组织中检测到NLR基因表达。研究表明正选择效应在NLRX1、NOD1、NLRC5基因作用明显,NLRC5在鸡回肠和肺组织中为表达活跃的功能基因。In order to further understand the characteristics of nucleotide-binding oligomerization domain-like receptors(NLRs) gene family in chicken,the chicken NLRs gene by using bioinformatics method through conserved domain search were identified,and the physicochemical properties,gene structure,phyletic relationship,positive selection,collinear relationship and expression of NLR gene were analyzed in this study.The results showed that a total of five chicken NLRs genes,named chNLRC5,chNLRC3,chNLRP3,chNLRX1 and chNOD1,were identified and distributed on five chromosomes,encoding 734 to 1 862 amino acids,and the relative molecular weight of the coding proteins ranged from 82 842.01 to 207 969.52 ku.The isoelectric point was 5.48 to8.64.chNLRC5,chNOD1 and chNLRP3 were acidic proteins,while chNLRP3 and chNLRX1 were alkaline proteins.Only chNOD1 was stable protein,and all five members were hydrophilic proteins.There were significant differences in gene structure,with 48,20,7,7 and 11 exons,respectively.The phylogenetic tree showed that the NLRs gene family was divided into five branches,and there was a strong collinear relationship among the NLR genes of chicken,mallard,turkey and Japanese quail.Thirteen positive selection sites were identified by selection pressure analysis of each member.NLR gene expression was detected in ileum and lung tissues of chicken.It′s concluded that positive selection effect had significant effects on NLRX1,NOD1 and NLRC5 genes,and NLRC5 was an active functional gene expressed in ileum and lung tissues of chicken.
