摘 要:目的探究甲状腺癌组织中微小RNA-93-5p(miR-93-5p)、乳腺癌转移抑制基因1相似基因(BRMS1L)表达及临床意义。方法选取行手术治疗的甲状腺癌患者150例,收集患者临床病理资料,术中取患者甲状腺癌组织及对应癌旁组织。利用癌症基因组图谱(TCGA)查询miR-93-5p、BRMS1L基因在甲状腺癌组织中的表达水平;采用免疫组织化学法检测甲状腺癌组织与癌旁组织中BRMS1L蛋白表达情况;采用qRT-PCR法分析甲状腺癌组织与癌旁组织中miR-93-5p表达;分析miR-93-5p、BRMS1L表达水平与甲状腺癌患者临床病理参数之间的关系;对患者随访3年,统计3年累计生存率,分析miR-93-5p、BRMS1L水平对患者3年累计生存率的影响。结果TCGA数据库显示,甲状腺癌组织中miR-93-5p基因表达水平与癌旁组织比较,差异无统计学意义(t=1.73,P>0.05);甲状腺癌组织中BRMS1L基因表达水平低于癌旁组织,差异有统计学意义(t=2.98,P<0.05);免疫组织化学染色结果显示,BRMS1L蛋白在甲状腺癌组织中的阳性表达率明显低于癌旁组织(χ^(2)=47.17,P<0.05),miR-93-5p在甲状腺癌组织中的表达水平明显高于癌旁组织(t=26.31,P<0.05)。miR-93-5p、BRMS1L表达水平与患者年龄、TNM分期、淋巴结转移和分化程度相关(χ^(2)分别=9.98、5.18;9.29、3.90;72.44、11.96;24.22、47.55,P均<0.05)。甲状腺癌组织中miR-93-5p高表达组和低表达组患者3年累计生存率分别为65.70%、27.00%,差异有统计学意义(χ^(2)=7.95,P<0.05);BRMS1L高表达组和低表达组患者3年累计生存率分别为31.80%、52.00%,差异有统计学意义(χ^(2)=3.98,P<0.05)。结论miR-93-5p在甲状腺癌组织中的表达水平高于癌旁组织,BRMS1L表达低于癌旁组织,均与患者年龄、TNM分期、淋巴结转移和分化程度相关,对患者3年累计生存率有一定的影响。Objective To explore the expression and clinical significance of microRNA-93-5p(miR-93-5p)and breast cancer metastasis suppressor 1 like(BRMS1L)in thyroid cancer tissues.Methods A total of 150 cases of thyroid cancer patients who underwent surgical treatment were selected.The clinicopathological data of patients were collected,and the patient's thyroid cancer tissue and corresponding adjacent tissues were taken during the operation.The cancer genome atlas(TCGA)was used to query the expression levels of miR-93-5p and BRMS1L genes in thyroid cancer tissues.Immunohistochemical method was used to detect the expression of BRMS1L protein in thyroid cancer tissues and adjacent tissues.The qRT-PCR was used to analyze the expression of miR-93-5p in thyroid cancer tissues and adjacent tissues.The relationship between the expression levels of miR-93-5p,BRMS1L and clinicopathological parameters in patients with thyroid cancer was analyzed.The patients were followed up for 3 years,the survival rate within 3 years was counted,and the effects of miR-93-5p and BRMS1L levels on the survival rate of patients within 3 years were analyzed.Results The TCGA database showed that the expression level of miR-93-5p gene in thyroid cancer tissue was higher than that in adjacent tissues,but the difference was not statistically significant(t=1.73,P>0.05).The expression level of BRMS1L gene in thyroid cancer tissue was lower than that in adjacent tissues(t=2.98,P<0.05).The results of immunohistochemical staining showed that the positive expression rate of BRMS1L protein in thyroid cancer tissues was significantly lower than that in adjacent tissues(χ^(2)=47.17,P<0.05),and the expression level of miR-93-5p in thyroid cancer tissues was significantly higher than that in adjacent tissues(t=26.31,P<0.05).The expression levels of miR-93-5p and BRMS1L were correlated with age,TNM stage,lymph node metastasis and differentiation degree(χ^(2)=9.98,5.18,9.29,3.90,72.44,11.96,24.22,47.55,P<0.05).The 3-year survival rate of patients in the miR-93-5p hi
