应用锌缺乏大鼠模型探索锌营养状况的评价指标

摘  要:目的通过使用缺锌饲料喂养大鼠,建立锌缺乏动物模型。探索不同指标评价锌缺乏的灵敏性和特异性及不同指标之间的联合应用。方法120只初断乳SPF级Wistar雄性大鼠应用Excel软件随机分为急性缺锌组、边缘性缺锌组、配饲组和对照组,每组30只大鼠。急性缺锌组饲喂急性缺锌饲料(锌含量1 mg/kg),边缘性缺锌组喂饲边缘缺锌饲料(锌含量4 mg/kg),配饲组和对照组喂饲AIN-93G饲料。除配饲组外其他各组大鼠自由饮食,配饲组大鼠的饲喂量依据急性缺锌组大鼠的摄食量而定。共喂养5周,记录大鼠摄食量、体重。实验第4天、第3周和第5周采血,采用质谱法测定血清锌,双抗体夹心法测定血清5’核苷酸酶、血清碱性磷酸酶、血清胸腺素、血清金属硫蛋白和血清血管紧张素转化酶含量。结果试验第4天时,急性缺锌组血清锌(1195.21μg/L)显著低于其余3组(P<0.001),且随喂养时间增加显著下降(P=0.0002);边缘性缺锌组大鼠血清锌(991.38μg/L)从第3周开始显著低于配饲组和对照组(P<0.05),但随着喂养时间增加血清锌无明显趋势变化。急性缺锌组5’核苷酸酶含量随喂养时间增加显著下降(P=0.0001),第5周时急性缺锌组5’核苷酸酶(41.43 ng/L)显著低于其他3组(P=0.0396)。急性缺锌组血清胸腺素含量随喂养时间增加显著下降(P=0.0355),第3周开始血清胸腺素(1467.66 ng/mL)显著低于配饲组和对照组(P=0.0006);边缘性缺锌组大鼠第5周血清胸腺素(1590.32 ng/mL)显著低于对照组(P<0.05)。血清碱性磷酸酶、血清金属硫蛋白和血清血管紧张素转化酶各组之间均无显著变化。急性缺锌组血清锌与血清5’核苷酸酶呈正相关关系(P=0.01);血清锌与血清胸腺素,血清胸腺素与血清5’核苷酸酶均无相关性。结论血清锌在急性锌缺乏时反应灵敏,但在边缘性锌缺乏时反应并不灵敏。血清胸腺素和血清5’核苷酸酶可能是急性锌缺乏�OBJECTIVE The animal model of zinc deficiency was established by feeding the rats with zinc deficiency diet.To explore the sensitivity and specificity of different indicators that evaluated zinc deficiency,and to explore the combined application of different indicators.METHODS A total of 120 weaned SPF Wistar male rats were randomly divided into four groups of thirty animals:acute zinc deficiency group,marginal zinc deficiency group,pair-fed group and control group.Rats in each group were fed the corresponding diet.Rats in acute zinc deficiency group were fed acute zinc deficiency diet(zinc content:1 mg/kg),rats in marginal zinc deficiency group were fed marginal zinc deficiency diet(zinc content:4 mg/kg),and rats in pair-fed group or control group were fed AIN-93 G diet.Except for the pair-fed group,the rats in other groups were fed ad libitum,and the feeding amount of the rats in the pair-fed group was determined according to the diet intake of the rats in the acute zinc deficiency group.Rats were fed for 5 weeks,and food intake and weight were recorded.Blood samples were collected on the 4 th day,the 3 rd week and the 5 th week to measure serum zinc using inductively coupled plasma-mass spectrometry(ICP-MS),to measure serum 5′nucleotide enzyme,serum alkaline phosphatase,serum thymosin,serum metallothionein and serum angiotensin converting enzyme with Elisa.RESULTS On the 4 th day,the serum zinc of the acute zinc deficiency group(1195.21μg/L)was significantly lower than that of the other three groups(P<0.001),and significantly decreased within the feeding time(P=0.0002);From the 3 rd week,the serum zinc of the marginal zinc deficiency group was significantly lower than that of the pair-fed group and control group(P<0.05),but there was no significantly change in the serum zinc as the feeding time increased.The 5′nucleotide enzyme of the acute zinc deficiency group were significantly decreased with the increasing feeding time(P=0.0001),and it was significantly lower than that of the other three groups in the
