摘 要:基于猪牛羊皮的特征肽段建立了一种鉴定皮革动物源属性的超高效液相色谱串联质谱法(UPLC-MS/MS)。皮革样品经高温高压提取蛋白,酶解消化,收集多肽待测溶液,经C_(18)柱分离,以0.1%甲酸水溶液和乙腈为流动相梯度洗脱,通过多反应监测(MRM)扫描,从多肽溶液中特征肽段的出峰情况来鉴定皮革样品的动物源属性。所建立的方法特异性强,适用范围广,准确可靠,为皮革鉴定提供了新的技术手段。An ultra high performance liquid chromatography tandem-mass spectrometry(UPLC-MS/MS)method was developed for the identification of animal origins for leather based on the peptide markers of porcine,bovine and sheep skins.Proteins were firstly extracted from the leather samples under high temperature and high pressure,and then digested by enzymolysis.The peptide solutions were separated by C_(18)column and eluted with the aqueous solution of formic acid and acetonitrile as mobile phases.The animal origins of the leather samples were identified by the peaks of peptide markers in the peptide solution by multiple reaction monitoring(MRM)scanning.The established method has strong specificity,wide application range,good accurateness and reliablity,and provides a new technical means for leather identification.
