摘　　要：目的:运用生物信息学技术分析淋巴增强因子-1(Lymphoid enhancer factor-1,LEF1)在胃癌中的表达及意义。方法:通过Oncomine、Ualcan、肿瘤免疫评估资源(TIMER)数据库检索并分析LEF1基因在胃癌中的表达。TIMER数据库分析LEF1在免疫浸润物中的表达;Ualcan数据库、Kaplan-Meier Plotter分析LEF1表达与患者生存及预后相关性。结果:Oncomine、TIMER、Ualcan数据库中均显示LEF1在胃癌中高表达,Kaplan-Meier Plotter分析得出LEF1高表达的胃癌患者无疾病生存时间、总生存时间较短(P<0.05);TIMER数据库分析显示在免疫微环境中高表达LEF1 mRNA巨噬细胞的胃癌患者5年生存预后较好,LEF1 mRNA在胃癌免疫微环境中的表达与多种免疫细胞在胃癌组织的免疫微环境中的表达明显相关。结论:LEF1在胃癌中表达增加,与胃癌患者不良预后相关,是胃癌预后的潜在的评判指标。Objective:To investigate the expression and significance of lymphoid enhancer factor-1(LEF1)in gastric cancer by bioinformatics.Methods:Oncomine,Ualcan and TIMER databases were used to search and analyze the expression of LEF1 gene in gastric cancer.TIMER database was used to analyze the expression of LEF1 in immune infiltrates,the correlation between LEF1 expression with patient survival and prognosis was analyzed by Ualcan database and Kaplan Meier Plotter.Results:LEF1 was highly expressed in Oncomine database,TIMER database and Ualcan database.Kaplan Meier Plotter shows that patients with high LEF1 expression have shorter disease-free survival time and overall survival time(P<0.05),TIMER database showed that macrophages with high LEF1 mRNA expression in the immune microenvironment have a better 5-year survival prognosis.LEF1 mRNA was significantly correlated with the expression of B cells,neutrophils,CD8+T cells,macrophages,CD4+T cells and dendritic cells in the immune microenvironment of gastric cancer(P<0.05).Conclusion:The increased expression of LEF1 in gastric cancer is associated with poor prognosis of gastric cancer patients,and it is a potential prognostic indicator of gastric cancer.
