摘 要:为了优选小桐子枝叶中生物碱的提取工艺,本文以小桐子中生物碱总含量为评价指标,通过单因素试验结合Box-Behnken响应面法,研究提取时间、提取溶剂体积分数、提取温度等因素对小桐子枝叶生物碱总含量的影响,优化选取最可行的提取工艺方法,并且进行验证试验。经试验得出最佳的提取工艺为提取时间20.15 min,提取溶剂体积分数为79.15%,温度为40℃。验证试验中生物碱总含量平均值为0.6011%(RSD=0.85%),与模型预测值的相对误差为0.59%,在允许的误差范围内,表明优选的工艺操作简便、效率高、稳定可靠,适用于小桐子枝叶中生物碱类成分的提取。In order to optimize the extraction process of alkaloids from the branches and leaves of Jatropha curcas L.,taking the total content in Jatropha curcas L.as the evaluation index,the effects of extraction time,volume fraction of extraction solvent and extraction temperature on the total content of alkaloids in the branches and leaves of Jatropha curcas L.were studied by single factor test combined with box-behnken response surface methodology,the most feasible extraction process was optimized and verified.The optimum extraction process was as follows:the extraction time was 20.15 min,the volume fraction of extraction solvent was 79.15%and the temperature was 40℃.In the validation test,the average value of the total alkaloid content was 0.6011%(RSD=0.85%),and the relative error between the total alkaloid content and the predicted value of the model was 0.59%,which was within the allowable error range.The results show that the optimized process is simple,efficient,stable and reliable,and is suitable for the extraction of alkaloids from the branches and leaves of Jatropha curcas L..
