摘  要:目的 对2016年云南省埃可病毒7型(echovirus 7,E-7)的分离株K1641/YN/CHN/2016(简称K1641)进行全基因组序列测定并分析其分子变异和遗传进化特性。方法 利用KMB17细胞分离病毒,提取病毒RNA,进行RT-PCR,并测序拼接获得全基因组序列。利用Mega 5、Geneious Basic 5.4.1和SimPlot 3.5.1等软件分析全基因组序列。结果 获得的K1641株全基因组序列长度为7 428 bp,编码含2 193个氨基酸的多聚蛋白。K1641全基因组核苷酸和氨基酸同源性与中国分离株40/Longyou/ZJ最为同源,分别为95.9%和98.9%;而与其他中国E-7流行株VP1的核苷酸和氨基酸同源性分别为95.1%~99.7%和82.5%~99.3%;与原型株VP1的核苷酸和氨基酸同源性为78.4%和93.1%。K1641与其他E-7毒株的各区段核苷酸一致性和氨基酸同源性分别为73.5%~97.3%和85.3%~100.0%,K1641与40/Longyou/ZJ的各区段核苷酸一致性和氨基酸同源性均超过90.0%,并且,氨基酸的同源性要高于核苷酸一致性。基于E-7全长VP1序列的种系进化分析,可分为8个簇,本次分离株K1641与2013中国分离株40/Longyou/ZJ关系较近,属于进化树上的一个分支。进行相似性分析,在位置350~600,与E-6/RO-24-9-79(LS451294)显示出99%的高相似性;其他位置与中国分离株40/Longyou/ZJ有较高(>92%)的相似性。结论 K1641分离株为E-7,与E-6/RO-24-9-79株可能存在重组事件发生,对E-7遗传特性研究具有参考意义。Objective To sequence the whole-genome of echovirus 7(E-7) strain of K1641/YN/CHN/2016(K1641) and to analyze the molecular variation and evolutionary characteristics. Methods K1641 was isolated from KMB17 cells and viral RNA was extracted. After RT-PCR amplification, the whole genome was sequenced and was analyzed using Mega 5,Geneious Basic 5.4.1 and Simplot 3.5.1. Results The whole genome of strain K1641 was 7 428 bp in length and encoded a polymeric protein containing 2 193 amino acids. The nucleotide and amino acid homologies of K1641 were 95.9% and 98.9%,respectively, with Chinese isolate 40/Longyou/ZJ.The nucleotide and amino acid homologies were 95.1%-99.7% and 82.5%-99.3% with other VP1 sequences of E-7 strains, and were 78.4% and 93.1% with VP1 of prototype strain. The nucleotide consistency and amino acid homology between K1641 and other E-7 strains were 73.5%-97.3% and 85.3%—100.0%,respectively. The nucleotide homology between K1641 and 40/Longyon/ZJ strain was more than 90.0%,and the amino acid homology was higher than that of nucleotide.Based on the phylogenetic analysis of E-7 full-length VP1 sequence, K1641 could be divided into 8 clusters. The strain K1641 isolated in Yunnan was clustered in the same of branch with 2013 Chinese strain 40/Longyou/ZJ. A similarity analysis showed a 99% similarity with E-6/RO-24-9-79(LS451294) at the position of 350—600 and over 92% similarity with Chinese isolates 40/Longyou/ZJ at other positions. Conclusions K1641 is defined as E-7, which might be recombined with E-6/Ro-24-9-79 strain,and K1641 has a high similarity with 40/Longyou/ZJ strain(>92%).Therefore, tt is important to study the genetic characteristics of E-7.
