摘  要:目的:分析环状RNA(circRNA)在宫颈癌组织及癌旁正常组织中的差异表达谱,探讨circRNA与宫颈癌发病机制及预后的关系。方法:收集我院2014年7-10月确诊为宫颈癌并行手术治疗的10例患者的癌组织及对应癌旁正常组织,采用基因芯片技术筛选两组样本间差异表达circRNA,随机选择差异表达circRNAs行实时荧光定量PCR检测(RT-qPCR)验证测序结果,选择RT-qPCR验证结果中表达差异最大的circRNA,结合癌症基因组图谱(TCGA)数据库分析其相对表达量与宫颈癌患者术后生存时间的关系,通过生物信息学方法分析差异表达circRNA宿主基因的生物学功能及信号通路。结果:宫颈癌组织及配对癌旁正常组织中发现DE circRNA有25749个(差异倍数≥2,P<0.05),其中表达上调的有12806个,表达下调有12943个;选取14个显著差异表达的circRNA(差异倍数≥3,P<0.05),RT-qPCR验证显示78.6%(11/14)与芯片结果一致;RT-qPCR验证结果显示hsa_circ_0008517在宫颈癌组织中显著低表达,且hsa_circ_0008517低表达组的中位生存时间低于高表达组,差异有统计学意义(P<0.05);GO富集分析显示异常表达circRNA参与细胞增殖、分化、信号转导等多种生物学过程和信号通路。结论:circRNA在宫颈癌组织中差异表达,与宫颈癌患者的预后密切相关,为宫颈癌的临床诊疗及评估预后提供新方向。Objective:To analyze the differential expression profiles of circRNA in cervical cancer and adjacent normal tissues,and to explore the relationship between circRNA and the pathogenesis and prognosis of cervical cancer.Methods:The cervical cancer and corresponding normal tissues of 10 patients were collected from July 2014 to October 2014 in our hospital.The differentially expressed circRNA were screened by gene chip technology.RT-qPCR was used to verify the sequencing results.The relationship between the relative expression and the survival time of cervical cancer was analyzed by TCGA database.The biological function and signal pathway of host genes for differentially expressed circRNA were analyzed by bioinformatics method.Results:25749 circRNA were found in cervical cancer tissues and matched normal tissues(multiple difference≥2,P<0.05),including 12806 up-regulated and 12943 down-regulated.14 significantly differentially expressed circRNAs were selected(multiple difference≥3,P<0.05),and 78.6%were confirmed by RT-qPCR.RT-qPCR results showed that hsa_circ_0008517 was significantly lower in cervical cancer,and the median survival time of hsa_circ_0008517 with low expression group was lower than that of high expression group,which the difference was statistically significant(P<0.05).GO enrichment analysis showed that abnormal expression of circRNA was involved in cell proliferation,differentiation,signal transduction and other biological processes and signal pathways.Conclusion:The differential expression of circRNA in cervical cancer is closely related to the prognosis of patients with cervical cancer,which provides a new direction for clinical diagnosis and treatment of cervical cancer and evaluation of prognosis.
