摘  要:目的口腔鳞癌(oral squamous cell carcinoma,OSCC)及正常组织的差异表达谱进行全基因组分析。方法采用全基因组表达谱芯片检测3例OSCC及正常组织的转录本,确定OSCC的lncRNA和mRNA差异表达谱。通过构建差异表达基因的lncRNA-mRNA共表达网络,挖掘网络节点基因。利用GO及KEGG富集分析,探究差异表达lncRNA潜在的生物学功能。通过cis和trans调控角度,预测差异表达lncRNA潜在的靶基因。选取部分差异表达lncRNA,进行qRT-PCR验证。结果芯片筛选出差异表达lncRNA 2294条和mRNA 1938条;lncRNA TMPRSS11BNL、lnc-WRN-10∶1是共表达网络的节点基因;GO分析显示,差异表达mRNA富集度的前三GO term是interleukin-1 binding、response to interferon-alpha和establishment of epithelial cell apical/basal polarity;KEGG分析显示,差异表达mRNA主要富集在38种生物学途径,包括许多癌症相关通路;靶基因预测表明,1470条差异表达lncRNA具有cis或trans调控靶基因的潜能;qRT-PCR验证与芯片结果相一致。结论lncRNA在OSCC与正常组织中具有差异表达,节点lncRNA是OSCC发病的潜在关键基因。Objective To investigate the differential expression profiles of oral squamous cell carcinoma(OSCC)and normal tissues.Methods Genome-wide expression spectrum chip was used to detect the transcripts of 3 cases of OSCC and normal tissues to determine the lncRNA and mRNA differentially expressed profiles.By constructing the lncRNA-mRNA co-expression network of differentially expressed genes,the hub genes were mined.The potential biological functions of differentially expressed lncRNAs were investigated by GO and KEGG enrichment analysis.The potential target genes of differentially expressed lncRNAs were predicted by the regulatory perspectives of cis and trans.Moreover some differentially expressed lncRNAs were selected for qRT-PCR validation.Results A total of 2,294 differentially expressed lncRNAs and 1,938 differentially expressed mRNAs were screened by the chip.LncRNA TMPRSS11BNL and lnc-WRN-10∶1 were hub genes of the co-expression network.GO analysis showed that the first three GO terms of differentially expressed mRNA enrichment were interleukin-1 binding,response to interferon-alpha,and establishment of epithelial cell apical/basal polarity.KEGG analysis demonstrated that differentially expressed mRNAs were mainly enriched in 38 biological pathways,including numerous cancer-related pathways.Target gene prediction revealed that 1,470 differentially expressed lncRNAs had the potential to regulate target genes in cis or trans.The qRT-PCR validation was consistent with the chip.Conclusion LncRNAs are differentially expressed in OSCC and normal tissues,and hub lncRNAs are potential key genes in the pathogenesis of OSCC.
