摘  要:通过静态吸附-解吸实验对8种不同极性大孔树脂对灵芝菌丝体中水溶性抗氧化成分的吸附进行筛选,确定中等极性的HPD-400大孔树脂为最佳.对上样后的大孔树脂利用不同体积比的乙醇(30%、50%、70%、95%)进行梯度洗脱,对比不同极性洗脱液的抗氧化活性,发现30%的乙醇可以洗脱绝大部分的抗氧化活性物质.将HPD-400大孔树脂分离后得到的30%乙醇洗脱物与水提物的体外抗氧化活性EC_(50)进行对比,结果表明经大孔树脂的富集,提取液的DPPH·清除能力可提高1倍,ABTS^(+)·清除能力可提高3倍,但羟自由基的清除能力有所降低,说明大孔树脂可有效富集具有DPPH·和ABTS^(+)·清除能力的水溶性抗氧化成分.The adsorption of 8 different polar macroporous resins to the water-soluble antioxidant components in Ganoderma lucidum mycelium was screened by static adsorption-desorption experiments,and the medium-polarity HPD-400 macroporous resin was determined to be the best.Gradient elution was performed on the loaded macroporous resin with different volume ratios of ethanol(30%,50%,70%,95%).The antioxidant activity of eluates with different polarities was compared,and it was found that 30%ethanol can elute most of the antioxidant active substances.The antioxidant activity EC_(50) of 30%ethanol eluate separated by HPD-400 macroporous resin was compared with that of water extract in vitro.The result showed that after the enrichment of the microporous resin,the treatment can double the DPPH·scavenging ability of the extract.The scavenging ability of ABTS^(+)·was increased by 3 times,but the scavenging ability of hydroxyl radicals was reduced.It shows that the macroporous resin can effectively enrich and separate water-soluble antioxidant components with DPPH·and ABTS^(+)·scavenging capabilities.
