摘 要:目的 研究苦参提取物对大鼠防龋作用的有效性及安全性。方法 将36只SPF-SD大鼠随机分为6组,接种变形链球菌(streptococcus mutans,S.mutans)UA159,参照经典的大鼠龋病模型进行动物实验,分别用浓度为2g/L、4g/L、8g/L的苦参提取物、0.12%的洗必泰及无菌蒸馏水干预大鼠口腔3周,通过菌落计数、Keyes龋齿记分、激光共聚焦显微镜(confocallaser scanning microscope,CLSM)、大鼠脏器系数及颊黏膜HE染色,综合评估苦参提取物防龋的有效性及安全性。结果 8g/L苦参提取物组的S.mutans水平、光滑面及窝沟的各级龋齿记分与龋病模型组均有统计学差异(P<0.05);CLSM下8g/L苦参提取物组牙釉质连续,牙磨片渗入的荧光强度明显降低。各组大鼠的口腔黏膜组织学结构无明显差异,各用药组之间体重无统计学差异,但肝脏脏器系数略高于空白对照组。结论 8g/L苦参提取物可有效阻止龋病的发生和进展,并初步表明具有一定的生物安全性。Objective To evaluate the anti-caries effect and safety of sophora flavescens Ait extract in vivo.Methods 36 SPF-SD rats were randomly divided into 6 groups,inoculated with S.mutans UA159,treated with sophora flavescens Ait extract at the concentration of 2g/L, 4g/L and 8g/L, 0.12% chlorhexidine and sterile distilled water for 3 weeks respectively. Colony count,Keyes caries score,CLSM,rat organ coefficient and buccal mucosa HE staining were used to evaluate the efficacy and safety of sophora flavescens Ait extract. Results In vivo data showed that the level of S.mutans,and the caries score in the 8g/L-treated animals were significantly reduced(P<0.05). In the 8g/L sophora flavescens Ait extract group,the enamel was continuous and fluorescence intensity was reduced when viewed by CLSM. There is no no statistically significant in weight gain and organ coefficient, and no sign of harm to the mucosa, indicating that sophora flavescens Ait extract possessed good biocompatibility in vivo. Conclusion The extract of 8g/L sophora flavescens Ait extract can effectively reduce the incidence and severity of dental caries without affecting the overall health of rats.
