摘 要:以健康的泌乳期中国荷斯坦奶牛的乳腺上皮细胞(Dairy cow mammary epithelial cells,DCMECs)为实验模型,探究miR-9-5p对奶牛乳腺上皮细胞乳脂合成的影响。通过体外转染miR-9-5p mimics的方式处理奶牛乳腺上皮细胞。随后采用MTT方法测定细胞增殖能力,采用western blotting技术检测乳脂合成相关信号通路蛋白SREBP1的表达情况,再采用甘油三脂检测试剂盒检测奶牛乳腺上皮细胞中甘油三脂含量的变化。实验结果显示miR-9-5p对奶牛乳腺上皮细胞的增殖有明显的抑制作用,同时miR-9-5p对乳脂合成相关信号通路蛋白SREBP1的表达也有明显的抑制作用,且显著抑制甘油三脂的合成,说明miR-9-5p对奶牛乳腺上皮细胞乳脂的合成有着负调控作用。本实验试图从分子水平揭示中国荷斯坦奶牛乳品质差异的原因,进一步完善中国荷斯坦奶牛泌乳的分子机制。The dairy cow mammary epithelial cells(DCMECs)of Chinese Holstein cows was used the experimental cell model to explore the effect of miR-9-5p on the synthesis of triglyceride in mammary epithelial cells of dairy cows.Dairy cow mammary epithelial cells were treated by miR-9-5p mimics in vitro.Then the proliferation ability of the cell was determined by MTT,the protein expression of srebp1 was detected by western blotting,and the triglyceride content of dairy cows mammary epithelial cells was detected by the triglyceride kit.The results showed that miR-9-5p had obvious inhibitory effect on the proliferation of DCMECs,inhibited the expression of SREBP1 and the triglyceride synthesis in dairy cow mammary epithelial cells.
