摘  要:[目的]本文以黄芪为原料,沼泽红假单胞菌(Rhodopseudomonas palustris)为发酵菌种,研究发酵对黄芪甲苷含量的影响。[方法]在单因素试验的基础上,以发酵时间(A)、培养基初始pH值(B)、发酵温度(C)和菌种接种量(D)为4个因素,每个因素设3水平,采用L_(9)(3^(4))正交表,以黄芪甲苷含量为指标进行正交试验,获得最佳发酵条件。[结果]4个因子对黄芪发酵液中的黄芪甲苷含量均产生了极显著性的影响(P<0.01),根据F值水平影响黄芪甲苷含量的因素强弱顺序为菌种接种量>培养基初始pH值>发酵时间>发酵温度;最佳发酵条件是A2B1C2D3,即菌种接种量10%、培养基初始pH值6.6、发酵时间5 d和发酵温度30℃,在此条件下黄芪甲苷含量达到了22.88μg·mL^(-1),比未发酵黄芪中黄芪甲苷含量提高了63.9%。[结论]本研究利用沼泽红假单胞菌在一定条件下发酵黄芪可促进黄芪中黄芪甲苷的释放,这对高效利用黄芪以及进一步开发传统中药资源具有非常重要的意义。[Objective]The objective of current study was to investigate the effects different fermentation conditions of Rhodopseudomonas palustris on the content of AstragalosidⅣusing astragalus as research material.[Methods]The L_(9)(3^(4))orthogonal table was used and the content of astragalosideⅣwere measured to select the optimal conditions using fermentation time(A),initial pH of medium(B),fermentation temperature(C),and inoculum amount(D)as the four main factors,and three levels were adopted for each factor.[Results]The optimal condition combination of R.palustris fermented Astagulus was A2B1C2D3.Under mentioned condition,the content of astragalosideⅣin fermentation medium was reached up to 22.88μg·mL^(-1),which was 63.9%higher than that in the control group.[Conclusion]The results have shown that Astragalus fermented by R.palustris can improve the astragalosideⅣcontent.This study provided a reference to optimize the process condition of the R.palustris fermented Astragalus,and is of great significance for the effective use of Astragalus and the in-depth development of traditional Chinese medicine.
