摘 要:目的为提高克罗诺杆菌属检出率,本实验室经过研究显色培养基的作用机理,优化研制出克罗诺杆菌属显色培养基(SWM)的配方,并与DFI琼脂、科玛嘉和国内某品牌的同类产品进行性能比较。方法通过GB 4789.28—2013对几种显色培养基的特异性和灵敏度进行比较,以及对人工污染样品和实际样品的检测差异进行分析。结果克罗诺杆菌属在SWM呈现特有的颜色,特异性强,可避免假阳性菌的干扰。在SWM上克罗诺杆菌属生长良好,生长率达到0.97,人工污染样品检测中能容易区分目标菌和其他杂菌,且可达到10 cfu的检出限。结论经过配方优化后的SWM对目标菌检测灵敏度更高,特异性更强,更易观察,综合检测效果可达到进口科玛嘉同类产品水平,避免了变形杆菌假阳性的缺点,可满足检测要求。Objective In order to improve the detection rate of Cronobacter sakazakii,this laboratory optimized the formulation of Cronobacter sakazakii chromogenic medium(SWM)by studying the mechanism of chromogenic medium,and compared its performance with DFI AGAR,French Comaga and a domestic brand.Methods The specificity and sensitivity of several chromogenic media were compared according to GB 4789.28—2013,and the detection differences between artificially contaminated samples and actual samples were analyzed.Results Cronobacter sakazakii present unique color on the SWM agar,with high specificity,and can avoid the interference of false positive.The growth rate of Cronobacter sakazakiion SWM reaches 0.97,it can be distinguished easily between target bacteria and other miscellaneous bacteria,and the detection limit reaches 10 cfu.Conclusion The optimized SWM has higher sensitivity,stronger specificity and easier observation for the detection of target bacteria.The comprehensive detection effect reaches the level of similar products imported,avoiding the false positive of proteus and meeting the detection requirements.
