摘  要:目的分析自噬相关基因在膀胱癌中表达模式并基于自噬相关基因构建膀胱癌预后模型。方法基于癌症基因组图谱数据库中膀胱癌患者基因表达谱数据和临床相关信息,分析自噬相关基因在膀胱癌中的表达模式;利用单因素、多因素Cox回归模型,筛选同膀胱癌预后相关的自噬相关差异表达基因,建立预后风险模型;利用不同来源的数据集(GSE48075)评估建立的预后风险模型。结果共筛选出38个膀胱癌组织中差异表达的自噬相关基因,MYC、SPHK1、NAMPT、P4HB、SPNS1、DIRAS3、TP63、APOL1、ITGA3等自噬相关基因表达同膀胱癌预后相关,利用MYC、SPHK1、NAMPT等自噬相关基因成功构建了膀胱癌预后风险模型,风险模型验证结果表明该模型对膀胱癌预后有较好的预测效果,表现为预后模型低风险组预后良好,高风险组预后较差。结论膀胱癌风险预后模型可较好地预测膀胱癌患者生存情况。Objective To analyze the expression pattern of autophagy-related genes in bladder cancer and construct a prognostic model based on these differentially expressed autophagy-related genes.Methods Gene expression profile data and clinical related information on bladder cancer patients were collected from the TCGA database.The differentially expressed autophagy related genes were filtered and used to evaluate their prognostic value with edgeR and survival packages.Based on these screened genes,a prognostic model was established with a multi-variate Cox regression analysis.Test data(GSE48075)was applied to validate the predictive ability of this prognostic model.Results 38 autophagy-related genes were found to be differentially expressed in bladder cancer tissues.9 autophagy-related genes(MYC,SPHK1,NAMPT,P4HB,SPNS1,DIRAS3,TP63,APOL1,ITGA3)were expressive in the bladder cancer prognosis.Based on these genes,a prognostic model was established,which showed that the low-risk group had a longer survival time than the highrisk group.Conclusion This study found that 38 autophagy-related genes were differentially expressed in bladder cancer tissues.9 autophagy-related genes could be used to establish a bladder cancer prognostic model,which could well predict the bladder cancer survival.
