摘　　要：目的:研究水调节蛋白(Aquaporin-1)在雷丸蛋白抑制胃癌细胞SGC-7901迁移中的作用。方法:以30μg/mL、60μg/mL、90μg/mL 3个浓度雷丸蛋白pPe Op干预SGC-7901细胞,以5-氟尿嘧啶(5-Fu)为阳性对照,乙酰唑胺与卡巴胆碱作为AQP1激动剂和抑制剂。细胞活力测定四氮唑蓝法(MTS)检测pPe Op对SGC-7901细胞活性影响;划痕法检测细胞迁移能力;Western blot检测AQP1蛋白表达情况。结果:加药组细胞存活率低于正常组,与pPe Op浓度呈负相关(P<0.05);乙酰唑胺与卡巴胆碱作用后细胞存活率无明显区别;划痕法结果显示胃癌细胞的迁移能力随pPe Op作用浓度的增加而减弱;Western blot结果显示,p Pe Op处理组AQP1表达含量呈下降趋势。高浓度乙酰唑胺作用后,AQP1表达量减少;较高浓度卡巴胆碱作用后,AQP1表达量增加。结论:雷丸蛋白pPe Op对胃癌SGC-7901细胞中水通道蛋白AQP1的表达和迁移具有显著的抑制作用。Objective:To study the role of water regulatory protein(AQP1) in the inhibitory effect of Omphalia lapidescens protein on the migration of gastric cancer cell line SGC-7901. Methods: SGC-7901 cells were intervened with three concentrations of Omphalia lapidescens protein pPe Op at 30 μg/mL、60 μg/mL、90 μg/mL. 5-fluorouracil(5-Fu) was used as positive control,acetazolamide and carbachol were used as AQP1 agonists and inhibitors. The effect of pPe Op on the activity of SGC-7901 cells was detected by MTS(3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium). Cell migration was detected by scratch method.The expression of AQP1 protein was detected by Western blot. Results: The cell survival rate in the dosing group was lower than that in the normal group, which was negatively correlated with the concentration of pPe Op(P<0.05).There was no significant difference in cell survival between acetazolamide and carbachol. The results of scratch method showed that the migration ability of gastric cancer cells decreased with the increase of pPe Op concentration;Western blot showed that the expression of AQP1 decreased in p Pe Op treatment group. The expression of AQP1 decreased after high concentration acetazolamide. The expression of AQP1 increased after higher concentration of carbachol. Conclusion:Omphalia lapidescens protein pPe Op can significantly inhibit the expression and migration of aquaporin AQP1 in gastric cancer SGC-7901 cells.
