摘  要:在抗肿瘤免疫治疗中,负责直接杀灭肿瘤细胞的免疫杀伤细胞如细胞毒性T淋巴细胞(cytotoxic T lymphocyte,CTL)发生功能紊乱是免疫疗法失效的直接原因。当宿主免疫反应失败且抗原长期存在时,抗原特异性CD8+T淋巴细胞转变为耗竭性T细胞。肿瘤中的T细胞耗竭可能与肿瘤微环境中富含的转化生长因子-β(transforming growth factor-β,TGF-β)存在潜在联系。目前已有多种针对TGF-β的抗肿瘤免疫策略,这些靶向TGF-β的治疗策略表现出良好的抗肿瘤功能,其中部分作用可能与改善T细胞的耗竭有关。T细胞耗竭也常常发生在慢性病毒感染中,共表达抑制性受体是耗竭性CD8+T细胞的关键特征。文章综述了肿瘤微环境及慢性病毒感染中TGF-β与T细胞耗竭之间潜在的联系和作用机制。In anti-tumor immunotherapy,the dysfunction of immune killer cells responsible for directly killing tumor cells,such as cytotoxic T lymphocytes(CTL),is the direct cause of the failure of immunotherapy.Antigen-specific CD8+T lymphocytes transform into exhausted T cells when the host immune response fails and antigens are present for a long time.The exhaustion of T cells in the tumor may be related to the abundant transforming growth factor-β(TGF-β)in the tumor microenvironment.There have been several anti-tumor immunological strategies targeting TGF-β,and these TGF-β-targeting therapeutic strategies show good anti-tumor activities,which may be partially related to the improvement of T cell exhaustion.T cell exhaustion also occurs during chronic viral infection,and co-expression of inhibitory receptors is a key feature of exhausted CD8+T cells.Here we reviewed the potential relationships and mechanisms between TGF-βand CTL exhaustion in tumor microenvironment and chronic viral infection.
