摘　　要：建立了养生酒中葛根素的高效液相色谱测定方法。不同葛根素含量的养生酒样品经适当稀释或水浴挥干、定容后,以甲醇-0.1%乙酸水溶液(梯度洗脱)为流动相,JADE-PAKODS-AQ C18色谱柱(4.6 mm×250 mm,5μm)分离的高效液相色谱法测定葛根素含量。结果显示:该方法在1.0~120 mg/L范围内线性关系良好,相关系数R2为0.9999,线性方程为y=45.849x-6.1179,方法检出限、定量限分别为0.10 mg/kg、0.40 mg/kg;仪器精密度良好,峰面积RSD为0.22%(n=6);养生酒中3个加标水平下的平均回收率范围为90.52%~93.98%(n=6),相对标准偏差为0.57%~2.37%。该方法灵敏度高、分析时间短、稳定性好、准确可靠,是测定葛根养生酒中葛根素含量的有效方法。In this study we established a method for the determination of puerarin content in Pueraria liqueur by HPLC.The experi-mental samples with different concentration levels of puerarin were measured after dilution or drying by water bath and dissolving with water.The methanol-0.1%acetic acid aqueous solution was used for gradient elution,and the JADE-PAK ODS-AQ C18 column(4.6 mm×250 mm,5μm)was used for the determination of puerarin content by HPLC.The results showed that the method had a good linear relationship in the range of 1.0 mg/L to 120 mg/L,with the correlation coefficient of 0.9999(y=45.849x-6.1179).The limit of detection was 0.10 mg/kg,and the limit of quantification was 0.40 mg/kg.The instrument had good precision.The RSD value of the peak area was 0.22%(n=6).The recoveries of the three spiking levels ranged from 90.52%to 93.98%(n=6).The RSD values of puerarin content ranged from 0.57%to 2.37%.The method has the advantages of high sensitivity,short analysis time,high stabili-ty,and high reliability,and can be used to determine the content of puerarin in Pueraria liqueur.
