摘  要:目的探讨长链非编码RNA类固醇受体RNA激活因子1(LncRNA-SRA1)在食管鳞状细胞癌(ESCC)组织中的水平和临床意义。方法收集38对ESCC组织和配对的癌旁组织,应用基因芯片技术筛选ESCC组织中lncRNAs的差异表达谱;通过实时定量聚合酶链式反应(qPCR)检测ESCC组织的LncRNA-SRA1水平,分析LncRNA-SRA1水平与ESCC临床病理特征的关系。采用Kaplan-Meier法分析ESCC患者不同LncRNA-SRA1水平的总生存期(OS),Cox比例风险回归模型用于评估独立预后因素。结果基因芯片及qPCR结果均发现在ESCC组织中LncRNA-SRA1水平较癌旁组织下调(P<0.05)。LncRNA-SRA1水平与ESCC浸润深度、淋巴结转移和TNM分期有关(P<0.05)。与LncRNA-SRA1高水平的ESCC患者相比,LncRNA-SRA1低水平ESCC患者的OS更短(P<0.05)。结论LncRNA-SRA1在ESCC组织中显著低表达,其低表达可能促进ESCC浸润转移,从而导致患者预后差。LncRNA-SRA1可能是治疗ESCC侵袭转移的新靶点,该指标结合TNM分期可用于判断ESCC患者的预后。Objective To explore the level and clinical significance of long noncoding RNA-steroid receptor RNA activator 1(LncRNA-SRA1)in esophageal squamous cell carcinoma(ESCC)tissues.Methods Thirty-eight pairs of ESCC tissues and adjacent normal tissues were acquired.Differential expression profiles of lncRNAs in ESCC tissues were screened by microarray assay.LncRNA-SRA1 level in ESCC tissues was detected by real-time quantitative polymerase chain reaction(qPCR),and relationship between LncRNA-SRA1 level and clinicopathological features of ESCC was analyzed.Kaplan-Meier analysis was performed to evaluate the overall survival(OS)of ESCC patients with a different expression level of LncRNA-SRA1.The Cox proportional hazards regression model was used to assess the independent prognostic factors.Results The results of gene chip and qPCR showed that LncRNA-SRA1 was in a pattern of low expression in ESCC tissues(P<0.05).LncRNA-SRA1 level was closely related to the depth of ESCC invasion,lymph node metastasis and TNM stage(P<0.05).Compared with ESCC patients with high LncRNA-SRA1 level,ESCC patients with low LncRNA-SRA1 level had shorter OS(P<0.05).Conclusion Low LncRNA-SRA1 level suggested a higher rate of ESCC invasion and lymph node metastasis,resulting in poor prognosis of patients.LncRNA-SRA1 may be a new target for the treatment of invasion and metastasis of ESCC.Combined detecting the expressions of LncRNA-SRA1 and TNM staging can be used to evaluate the prognosis of ESCC patients.
