摘 要:目的探讨疏肝健脾法对肝郁脾虚型功能性消化不良(FD)大鼠胃窦组织中胃肠激素的影响。方法将60只SPF级雄性SD大鼠随机分为正常组、模型组、柴术理胃饮低剂量组、柴术理胃饮中剂量组、柴术理胃饮高剂量组和多潘立酮组,每组10只。除正常组外,其余组均采用复合因素方法制备肝郁脾虚型FD模型。造模成功后,柴术理胃饮低、中、高剂量组分别给予13.5 g/kg、27 g/kg、54 g/kg柴术理胃饮灌胃,多潘立酮组给予3.125 mg/kg多潘立酮溶液灌胃,正常组、模型组给予等量生理盐水灌胃,均1次/d,连续21 d。记录各组大鼠末次灌胃当天体重及进食量,计算各组大鼠末次灌胃后胃排空率和小肠推进率,HE染色观察胃窦病理形态变化,蛋白质印迹法检测胃窦组织中P物质(SP)、胃泌素(GAS)、胃动素(MTL)及血管活性肠肽(VIP)蛋白表达情况,Real Time PCR法检测胃窦组织中SP mRNA表达情况。结果模型组大鼠末次灌胃当天体重、进食量及末次灌胃后胃排空率、小肠推进率均明显低于模型组(P均<0.05),柴术理胃饮各组和多潘立酮组大鼠体重、进食量、胃排空率及小肠推进率均明显高于模型组(P均<0.05)。HE染色显示模型组大鼠胃窦黏膜层有轻度淋巴细胞浸润,柴术理胃饮各组和多潘立酮组大鼠胃窦黏膜层淋巴细胞浸润程度轻于模型组。与正常组比较,模型组大鼠胃窦组织中SP、GAS、MTL蛋白表达量及SP mRNA表达量均明显降低(P均<0.05),VIP蛋白表达量明显增高(P<0.05);柴术理胃饮中、高剂量组大鼠胃窦组织中SP、GAS、MTL蛋白表达量和柴术理胃饮各组、多潘立酮组SP mRNA表达量均明显高于模型组(P均<0.05),柴术理胃饮各组和多潘立酮组大鼠胃窦组织中VIP蛋白表达量均明显低于模型组(P均<0.05)。结论疏肝健脾法可以促进肝郁脾虚型FD大鼠胃肠蠕动,上调胃窦组织中SP、GAS、MTL表达和下调VIP表达可能是其治疗FD的重要�Objective It is to investigate the effect of soothing liver and fortifying spleen method on gastrointestinal hormones in gastric antrum of rats with functional dyspepsia(FD)of type of liver stagnation and spleen deficiency.Methods Sixty SPF male SD rats were randomly divided into normal group,model group,low-dose Chaizhu Liweiyin group,medium-dose Chai Zhu Liweiyin group,high-dose Chai Zhu Liweiyin group and domperidone group,10 rats in each group.Except for the normal group,the other groups were all used the compound factor method to prepare the FD model of liver stagnation and spleen deficiency.After successful modeling,the low-dose,medium-dose and high-dose groups of Chai Zhu Liweiyin were respectively given 13.5 g/kg,27 g/kg,54 g/kg Chai Zhu Liweiyin by gavage,and the domperidone group was given 3.125 mg/kg domperidone solution by gavage,the normal group and the model group were given the same amount of normal saline by gavage,all once a day,continuously treated for 21 days.The body weight and food intake of the rats in each group on the day of the last gavage were recorded,and the gastric emptying rate and the small intestine advancement rate after the last gavage were calculated.The pathological changes of gastric antrum was observed by HE staining,and the protein expression of P Substance(SP),gastrin(GAS),motilin(MTL)and vasoactive intestinal peptide(VIP)was detected by Western blotting,and SP mRNA expression in gastric antrum tissue was detected by Real Time PCR.Results The body weight,food intake on the day of the last gavage,gastric emptying rate and small intestine propulsion rate of the rats after the last gavage in the model group were significantly lower than those in the model group(all P<0.05),the body weight,food intake,gastric emptying rate and small intestinal propulsion rate of the rats in each Chai Zhu Liweiyin group and domperidone group were significantly higher than those in the model group(all P<0.05).HE staining showed that there was mild lymphocyte infiltration in the gastric antrum muc
