摘 要:目的研究“培土生金法”早期干预对哮喘模型幼鼠肠道菌群的影响。方法将32只7日龄BALB/c雌性幼鼠随机分为空白组、模型组、异功散组、益生菌组,每组8只。除空白组外,其余组均以鸡卵清蛋白(OVA)致敏激发法制备哮喘幼鼠模型。从造模起始日开始,异功散组给予异功散煎液灌胃,益生菌组给予浓度为5×10^(8)CFU/mL的酪酸梭菌溶液灌胃,空白组和模型组给予PBS溶液灌胃,均连续灌胃18 d。实验结束后取各组幼鼠结肠粪便,以16S-rRNA扩增子高通量测序法检测肠道菌群。结果与空白组比较,模型组的拟杆菌门丰度减少,厚壁菌门丰度增加,肠道菌群丰度chao1指数及observed_species指数和菌群多样性shannon指数及simpson指数均有下降趋势,但差异均无统计学意义(P均>0.05)。与模型组比较,异功散组的拟杆菌门丰度增加,厚壁菌门丰度减少,肠道菌群的丰度指数chao1明显降低(P<0.05);益生菌组幼鼠肠道菌群丰度指数和菌群多样性指数均有增加趋势,但差异均无统计学意义(P均>0.05)。异功散组的优势菌属还包含螺杆菌属、黄酮菌属。结论“培土生金法”早期干预可改变哮喘模型幼鼠的肠道菌群,其防治哮喘作用可能与某些关键菌群如拟杆菌、螺杆菌、黄酮菌等相关。Objective It is to explore the effect of early intervention of“therapy for tonifying the spleen to help the lung”on the intestinal flora of young asthma model mice.Methods Thirty-two 7-day-old BALB/c female pups were randomly divided into blank group,model group,Yigong Powder group,and probiotics group,with 8 mice in each group.Except for the blank group,the mice in the other groups were sensitized with chicken ovalbumin(OVA)to establish asthmatic models.From the starting day of modeling,the Yigong Powder group was given Yigong Powder decoction by gavage,the probiotics group was given a solution of Clostridium butyricum with a concentration of 5×10^(8)CFU/mL by gavage,and the blank group and model group were given PBS solution by gavage,all the groups were treated for 18 days.After the experiment,the colonic feces of young mice in each group were collected,and the intestinal flora was detected by 16S-rRNA amplicon high-throughput sequencing method.Results Compared with the blank group,the abundance of Bacteroidetes in the model group decreased,and the abundance of Firmicutes increased,the chao1 index and observed_species index of intestinal flora abundance and the shannon index and simpson index of flora diversity all showed a downward trend,but the differences were not statistically significant(all P>0.05).Compared with the model group,the abundance of Bacteroidetes in Yigong Powder group increased,the abundance of Firmicutes decreased,and the abundance index chao1 of intestinal flora significantly decreased(P<0.05).The intestinal flora abundance index and flora diversity index of the young mice in the probiotic group both tended to increase,but the differences were not statistically significant(all P>0.05).The dominant bacterial genera in Yigong Powder group also included Helicobacter and Flavonoids.Conclusion The early intervention of“therapy for tonifying the spleen to help the lung”can change the intestinal flora of young asthma model mice,and its prevention and treatment of asthma may be related to
