摘 要:旨在分析两面神激酶2(Janus kinase 2,JAK2)和杆状病毒IPA重复序列蛋白6(Baculoviral inhibitors of apoptosis repeat-containing 6,BIRC6)基因的多态性与云上黑山羊产羔性能的关联性。利用Sequenom MassARRAY^(■)SNP技术对JAK2和BIRC6的7个候选位点进行分型,探索其在云上黑山羊(n=544)、济宁青山羊(JN,n=133)和辽宁绒山羊(LN,n=91)3个群体中的遗传学特征,并对其与云上黑山羊产羔性能(包括产羔数、初生窝重和断奶窝重)的关联性进行分析。结果表明,JAK2基因的g.39274131C>T和g.39301320T>C位点、BIRC6基因的g.14843779A>G和g.14903534A>G位点在不同繁殖力品种(JN和LN)中的基因型分布存在极显著差异(P<0.01),而BIRC6基因的g.14883749G>C、g.14886515C>G和g.14903556G>C位点在不同繁殖力品种(JN和LN)中的基因型分布无显著差异(P>0.05)。在云上黑山羊中,所有候选位点对产羔数、出生窝重和断奶窝重均无显著性影响(P>0.05)。综上,JAK2基因和BIRC6基因7个位点均不适合作为云上黑山羊产羔性能选育的分子标记。This study was performed to analyze the association between the Janus kinase 2(JAK2)and Baculoviral inhibitors of apoptosis repeat-containing 6(BIRC6)genes and kidding performance of Yunshang Black goats(YS).The Sequenom MassARRAY^(■)system was used for genotyping of the seven candidate loci of JAK2 and BIRC6,of which the genetic characteristics of YS(n=544),Jining Grey goats(JN,n=133)and Liaoning Cashmere goats(LN,n=91)were explored,and whose association with kidding performance including litter size,litter weight at birth and at weaning was also analyzed.The genotype distributions of candidate loci(JAK2 g.39274131C>T,JAK2 g.39301320T>C,BIRC6 g.14843779A>G,BIRC6 g.14903534A>G)in breeds with different fecundity(JN and LN)were extremely significantly different(P<0.01).However,the genotype distributions of candidate loci(BIRC6 g.14883749G>C,g.14886515C>G,g.14903556G>C)in breeds with different fecundity(JN and LN)were not significantly different(P>0.05).The association analyses of YS showed that there were no significant effects of all the loci on litter size,litter weight at birth and at weaning(P>0.05).These results suggested that the seven loci at JAK2 and BIRC6 were unsuitable as potential markers in YS selection and breeding.
