摘　　要：目的探讨lncRNA-KAT7在肺癌组织中表达水平变化及对肺癌细胞侵袭迁移的影响。方法采用实时荧光定量PCR(q-PCR)检测lncRNA-KAT7在肺癌组织中的表达情况,分析lncRNA-KAT7的表达水平与其临床病理特征的关系;采用瞬时转染技术在A549细胞内过表达lncRNA-KAT7,q-PCR检测lncRNA-KAT7的过表达效率;采用Transwell实验分析lncRNA-KAT7过表达对A549细胞迁移和侵袭能力的影响。结果肺癌组织中lncRNA-KAT7的表达水平明显低于癌旁组织(P<0.05);选取的38份肺癌组织及癌旁组织标本中,84.2%(32/38)的肺癌组织标本中lncRNA-KAT7的表达水平明显低于癌旁组织标本;lncRNA-KAT7的表达水平与患者的淋巴结转移(P=0.043)、肺癌组织学类型(P=0.001)及年龄(P=0.027)有关,但与患者的肿瘤分化程度、肿瘤最大径、TNM分期、T分级及性别无关(P>0.05);Transwell实验结果表明,与pcDNA-3.1组比较,pcDNA-lncRNA-KAT7组平均每个视野穿至小室外的迁移细胞数和侵袭细胞数均明显减少(P<0.05)。结论lncRNA-KAT7在肺癌的发生发展过程中具有调控肿瘤细胞迁移和侵袭能力的作用,其有望成为肺癌分子靶向治疗中的潜在靶点。Objective To investigate the expression level of lncRNA-KAT7 in lung cancer tissues and its effect on the invasion and migration of lung cancer cells.Methods Real-time quantitative PCR(q-PCR)was used to detect the expression of lncRNA-KAT7 in lung cancer tissues,and the relationship between the expression level of lncRNA-KAT7 and its clinicopathological characteristics was analyzed.The lncRNA-KAT7 was overexpressed in A549 cells by transient transfection technology,and the overexpression efficiency of lncRNA-KAT7 was detected by q-PCR.The effects of lncRNA-KAT7 on the migration and invasion ability of A549 cells were analyzed by Transwell assay.Results The expression level of lncRNA-KAT7 in lung cancer tissues was significantly lower than that in adjacent tissues(P<0.05).Among the selected 38 lung cancer tissue and paracancerous tissue samples,the expression level of lncRNA-KAT7 in 84.2%(32/38)of lung cancer tissue samples were significantly lower than that in adjacent tissue samples.The expression level of lncRNA-KAT7 was related to the lymph node metastasis(P=0.043),histological type(P=0.001)and the age of patients(P=0.027),but it was not related to the degree of tumor differentiation,tumor size,TNM stage,T stage and gender of patients(P<0.05).The results of Transwell experiment showed that,compared with the pcDNA-3.1 group,the average number of migrating cells and invasive cells per field in the pcDNA-lncRNA-KAT7 group were significantly reduced(P<0.05).Conclusion lncRNA-KAT7 has the function of regulating tumor cell migration and invasion in the occurrence and development of lung cancer,and is expected to become a potential target in molecular targeted therapy of lung cancer.
