归脾丸对小鼠抑郁行为及单胺递质的影响

摘　　要：目的:探讨归脾丸对小鼠抑郁行为及单胺递质的影响。方法:将ICR小鼠随机分为空白组、模型组、阳性药组及归脾丸高、中、低剂量组各10只,均按20 mL/kg灌胃给药,每日1次,连续灌胃7天,利用利血平诱导小鼠抑郁模型,观察小鼠体温、眼睑下垂及强迫游泳实验(forced swimming test,FST)、悬尾实验(tail suspension test,TST)和旷场实验(open field test,OFT)相应指标变化情况;采用酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)法测定小鼠血浆5-羟色胺(5-hydroxytryptamine,5-HT)、去甲肾上腺素(norepinephrine,NE)、多巴胺(dopamine,DA)含量。结果:归脾丸高、中、低剂量能提高抑郁模型小鼠体温(P<0.001),改善眼睑下垂(P<0.05,P<0.01);归脾丸高剂量能缩短小鼠游泳不动时间(P<0.05),提高旷场实验总运动距离(P<0.01)及小鼠血浆5-HT(P<0.01)、NE(P<0.001)和DA(P<0.001)含量。结论:归脾丸的抗抑郁作用机制可能与调节单胺递质含量有关。Objective:To discuss the effects of Guipi pills on depression behaviors and monoamine neurotransmitter in mice.Methods:The ICR mice were randomized into the blank group,the model group,positive medicine group,high,moderate and low dose groups of Guipi pills,with ten mice in each group.all the groups were drenched with 20 mL/kg of herbs,once each day,gavage administration for seven consecutive days,the models were induced by reserpine.To observe body temperature,eyelid ptosis,the related indexes of forced swimming test(FST),tail suspension test(TST) and open field test(OFT) of the mice;ELISA method was used to detect the contents of plasma 5-HT,NE and DA.Results:High,moderate and low doses of Guipi pills could lift body temperature in depression mice model(P<0.001),and improve eyelid ptosis(P<0.05,P<0.01);high dose of Guipi pills could shorten swimming immobility time(P<0.05),raise total movement distance of OFT(P<0.01),lift the contents of plasma 5-HT(P<0.01),NE(P<0.001) and DA(P<0.001).Conclusion:Antidepressant mechanism of Guipi pills might be related to regulating the contents of monoamine neurotransmitter.
