摘 要:目的:观察五味子根、茎、叶、果多糖对D-半乳糖所致衰老小鼠运动耐力的影响。方法:将雄性ICR小鼠随机分为空白组、模型组、五味子根多糖组、茎多糖组、叶多糖组及果多糖组,分别给予蒸馏水及总糖含量为35 mg·kg^(-1)的根、茎、叶及果粗多糖灌胃,30 min后,除空白组皮下注射生理盐水外,其他组均皮下注射300 mg·kg^(-1)D-半乳糖,每日1次,持续6周。通过转棒实验、前肢握力实验和负重游泳实验评价抗疲劳能力;通过化学比色法检测小鼠血清中尿素氮(BUN)、乳酸(LD)、乳酸脱氢酶(LDH)、肌酸激酶(CK)、超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)和活性氧(ROS)等疲劳和氧化相关指标;用蛋白免疫印迹法(Western blot)检测小鼠骨骼肌细胞中促凋亡蛋白B细胞淋巴瘤-2(Bcl-2)相关X蛋白(Bax)、抗凋亡蛋白Bcl-2及活化型胱天蛋白酶-3(cleaved Caspase-3)的表达情况。结果:在疲劳转棒实验中,与空白组比较,模型组小鼠转棒停留时间显著缩短(P<0.01);与模型组比较,五味子根、茎及果多糖组小鼠转棒时间均显著增加(P<0.01);前肢握力实验,与空白组比较,模型组小鼠前肢握力显著缩短(P<0.01),与模型组比较,五味子根、茎、叶及果多糖组小鼠前肢握力显著增加(P<0.01);负重游泳实验,与空白组比较,模型组小鼠负重游泳时间显著缩短(P<0.01),与模型组比较,五味子根、茎、叶及果多糖组小鼠负重游泳时间显著延长(P<0.01);生化试剂盒检测显示,与空白组比较,模型组小鼠BUN、LD含量及LDH、CK活性显著升高(P<0.01);与模型组比较,五味子根、茎及果多糖组小鼠BUN含量及LDH活性明显下降(P<0.05,P<0.01),五味子根、茎、叶及果多糖组小鼠LD含量及CK活性明显下降(P<0.05,P<0.01);与空白组比较,模型组小鼠SOD、GSH-Px活性显著下降(P<0.01),MDA、ROS含量显著升高(P<0.01);与模型组比较,根、茎及果多糖组小鼠SOD活性Objective:To observe the effect of polysaccharides from root,stem,leaf and fruit of Schisandra chinensis on exercise endurance in the aging mice induced by D-galactose.Method:Male ICR mice were randomly assigned into six groups:blank control group,model group,root polysaccharide group,stem polysaccharide group,leaf polysaccharide group and fruit polysaccharide group.The mice were administrated with distilled water or root,stem,leaf and fruit polysaccharide(total sugar content of 35 mg·kg^(-1))by gavage.Thirty minutes after the administration,the blank control group was subcutaneously injected with normal saline,and the other groups with D-galactose(300 mg·kg^(-1)),once daily for 6 weeks.The anti-fatigue effects were evaluated by rotarod test,forelimb grip strength test,and weight-loaded swimming test.The fatigue and oxidation indicators such as blood urea nitrogen(BUN),serum lactic acid(LD),lactic dehydrogenase(LDH),creatine kinase(CK),superoxide dismutase(SOD),malondialdehyde(MDA),glutathione peroxidase(GSHPx),and reactive oxygen species(ROS)were measured by chemical colorimetry.The protein levels of proapoptotic protein B cell lymphoma-2(Bcl-2)-associated X protein(Bax),anti-apoptotic Bcl-2 and cleaved cysteinyl aspartate-specific protease-3(cleaved Caspase-3)in mouse skeletal muscle were detected by Western blot.Result:In the rotarod test,the time on rod was shorter in the model group than in the blank control group(P<0.01)and the root,stem and fruit polysaccharide groups(P<0.01).In the forelimb grip strength test,the forelimb grip strength in the model group was lower than that in the blank control group(P<0.01)and the root,stem,leaf and fruit polysaccharide groups(P<0.01).In the weight-loaded swimming test,the weight-loaded swimming time in the model group was shorter than that in the blank control group(P<0.01)and the root,stem,leaf and fruit polysaccharide groups(P<0.01).Compared with those in the blank control group,the BUN,LD,LDH and CK levels significantly increased in the model group(P<0.05,P<0.01).T
