摘  要:目的探讨性别决定区Y框蛋白2(SOX2)和程序性细胞死亡因子4(PDCD4)在正常脑组织和胶质瘤组织中的表达水平及临床意义。方法收集2016年5月至2021年10月大庆油田总医院(齐齐哈尔医学院附属第十医院)病理确诊的13例正常脑组织和70例胶质瘤组织,采用免疫组织化学方法检测SOX2和PDCD4在上述组织中表达,结合相关临床资料行χ^(2)检验相关性分析。结果胶质瘤组织中SOX2阳性表达率75.71%(53/70),明显高于正常脑组织中阳性表达率15.39%(2/13),两者差异有统计学意义(χ^(2)=17.851,P<0.01)。胶质瘤组织中PDCD4阳性表达率34.29%(24/70),明显低于正常脑组织中阳性表达率76.92%(10/13),两者差异有统计学意义(χ^(2)=8.242,P<0.01)。SOX2和PDCD4表达水平与胶质瘤组织学分级明显相关(χ^(2)=4.953、5.587,P<0.05)。SOX2和PDCD4表达水平与胶质瘤性别、年龄、肿瘤部位无明显相关(χ^(2)=0.026、0.064、0.071、0.021、0.018、0.095,P>0.05)。结论胶质瘤组织中SOX2高表达和PDCD4低表达与胶质瘤的恶性程度和不良预后密切相关,检测SOX2和PDCD4有助于预测可胶质瘤进展及预后。Objective To investigate the expression levels of sex determining region Y-box 2(SOX2)and programmed cell death 4(PDCD4)in normal brain and glioma tissues and their clinical significance.Methods A total of 13 normal brain tissues and 70 glioma tissues were collected from Daqing Oilfield General Hospital(10th Affiliated Hospital of Qiqihar Medical College)from May 2016 to October 2021.The expression levels of SOX2 and PDCD4 in the above tissues were detected by immunohistochemistry,and correlation analysis was performed by chi-square test combined with relevant clinical data.Results The positive expression rate of SOX2 in glioma tissue was 75.71%(53/70),which was significantly higher than that in normal brain tissue[15.39%(2/13)],and the difference was statistically significant(χ^(2)=17.851,P<0.01).The positive expression rate of PDCD4 in glioma tissue was 34.29%(24/70),which was significantly lower than that in normal brain tissue[76.92%(10/13)],and the difference was statistically significant(χ^(2)=8.242,P<0.05).The expression levels of SOX2 and PDCD4 were significantly correlated with histological grade of glioma(χ^(2)=4.953,5.587,P<0.05).There was no significant correlation between the expression levels of SOX2 and PDCD4 and glioma gender,age and tumor site(respectively,χ^(2)=0.026,0.064,0.071,0.021,0.018,0.095,P>0.05).Conclusion The high expression of SOX2 and low expression of PDCD4 in glioma tissues are closely related to the malignant degree and poor prognosis of glioma.SOX2 and PDCD4 may become markers for predicting the progression and prognosis of glioma.
