摘 要:目的:观察(0~30kPa, 1~4h)静水压力条件下髁突软骨细胞凋亡的变化,总结髁突软骨细胞在一定压力条件下的凋亡规律,为限定临床中有益于髁突的受压条件提供理论依据。方法:将大鼠髁突软骨细胞分别按照加压条件(静水压力0kPa、6kPa、8kPa、10kPa、20kPa、30kPa和时间1h、2h、4h)分成18组,其中0kPa为对照组,其余为实验组,按照每组对应的加压条件施加静水压力后收集细胞,以Western Blot方法检测各组细胞中凋亡相关基因FAS的蛋白表达量,重复3次,取平均值。结果:加压时间为1h、2h、4h时,6~10kPa实验组的FAS蛋白量均低于对照组,差异具有统计学意义(P<0.05);当压力值为20kPa/4h、30kPa/1h、30kPa/2h或4h时,实验组高于对照组,差异具有统计学意义(P<0.05);当压力为20kPa/1h、20kPa/2h时,实验组低于对照组,但差异无统计学意义(P>0.05)。结论:髁突受到较小压力时,压力对细胞的影响表现为抑制凋亡,有益于髁突的改建和生长;受到较大压力时表现为促进细胞凋亡,不利于髁突的健康;同时髁突受力的时间不要过长,长时间大压力对髁突改建和生长不利。Objective:To observe the apoptosis of mandibular condylar cartilage cells(MCCs)under different hydrostatic pressure(0~30 kPa, 1~4 h).Summed up the regularity of apoptosis of condylar chondrocytes under certain pressure conditions in order to provided theoretical basis of beneficial pressure conditions for condyle.Methods:Divided the MCCs into 18 groups randomly according to different pressure conditions(Hydrostatic pressure: 0 kPa, 6 kPa, 8 kPa, 10 kPa, 20 kPa, 30 kPa;Time: 1 h, 2 h, 4 h). 0 kPa was the control group and the others were experimental group.Brought corresponding pressure to each group, then collected the cells.The expression of FAS protein was detected by Western Blot in each group.Repeated 3 times and took the average.Results:The FAS protein in experimental group expressed less than control group with the pressure 6 kPa, 8 kPa, 10 kPa under the duration of 1 h, 2 h or 4 h, the difference is statistically significant(P<0.05).FAS protein in experimental group expressed much more than control group with 20 kPa/4 h, 30 kPa/1 h, 30 kPa/2 h, 30 kPa/4 h(P<0.05).When the pressure was 20 kPa/1 h, 20 kPa/2 h, the difference is not statistically significant between experimental group and control group(P>0.05).Conclusion:When the condyle is under small pressure, the effect of pressure on the cells is shown as inhibition of apoptosis, which is beneficial to the reconstruction and growth of condyle;when the condyle is under greater pressure, it can promote cell apoptosis, which is not conducive to the health of condyle;at the same time, the duration of condylar stress should not be too long, and long-term high pressure is not conducive to the remodeling and growth of condyle.
