摘 要:为探究南欧大戟(Euphorbia peplus L.)叶绿体基因组序列特征及系统进化位置关系。本研究以南欧大戟总DNA为研究对象,利用Illumina HiSeq 2500高通量测序仪测定南欧大戟的叶绿体基因组序列,采用GetOrganelle进行完整叶绿体基因组的组装,并借助生物信息学工具进行基因组的注释分析、图谱构建和系统发育分析。结果表明,南欧大戟叶绿体基因组全长159 466 bp,呈典型的四分体环状结构,包括一个大单拷贝区、一个小单拷贝区和两个反向互补重复区,其大小分别为89 398、17 802和26 133 bp,GC值为35.8%;共编码基因130个,其中包括8个rRNA基因、37个tRNA基因和85个蛋白编码基因。基于23种植物系统发育分析结果表明,南欧大戟与同亚科植物浆果乌桕的亲缘关系最近。本研究获取了南欧大戟的叶绿体基因组特征信息和系统发育位置,为该植物的资源开发、品种鉴定和遗传进化研究提供了新的材料基础。In order to explore the chloroplast genome sequence characteristics and phylogenetic relationship of Euphorbia peplus L..In this study,using total DNA of E.peplus as material,Illumina HiSeq 2500 high-through-put sequencer was used to determine the chloroplast genome of E.peplus,and the complete chloroplast genome was assembled by GetOrganelle.Bioinformatics tools were used to annotate,map and phylogenetic analysis of the genome.The results showed that the total length of the chloroplast genome of E.peplus was 159 466 bp,with a typical tetrad ring structure,including a large single copy region,a small single copy region and two inverted repeat regions,their sizes were 89 398,17 802 and 26 133 bp,respectively,and the GC value of 35.8%.A total of 130 genes were encoded,including 8 rRNA genes,37 tRNA genes and 85 protein genes.The phylogenetic analysis of 23 species showed that E.peplus had the closest relationship with Balakata baccata.In this study,the chloroplast genome characteristic information and phylogenetic location of E.peplus were obtained,which provided a new material basis for resource exploitation,variety identification and genetic evolution research of this plant.
