摘 要:文章以野葛藤茎及葛根为原材料,通过水蒸气蒸馏-乙醚萃取法提取葛藤茎挥发油成分,并用气相色谱-质谱联用(GC/MS)分析其挥发油成分。以总黄酮的得率为指标,芦丁作为对照样品,亚硝酸钠-硝酸铝法为黄酮类化合物分析方法,采用超声波辅助提取葛根总黄酮。通过单因素实验考察乙醇浓度、固液比、超声功率、提取时间、提取温度对得率的影响,并选用正交实验确定了葛根总黄酮的提取工艺。结果表明,从葛藤茎中成功分离并鉴定出10种主要挥发性成分,其占总成分的83.94%,其中醇类占49.68%,酯类占15.85%,烷类占5.81%,酚类占12.60%。其中蓝桉醇的含量最高,占31.30%。芦丁样品做出的标准曲线方程为Y=11.146X-0.006,R^(2)=0.9992。葛根提取物的精密度和稳定性实验的RSD分别是0.227%、1.23%,平均加标回收率达到98.6%,RSD为1.86%。葛根总黄酮最佳提取工艺为提取温度80℃、乙醇浓度40%、提取时间20 min、固液比1∶50(g/mL)、超声输出功率300 W。在此条件下提取1次,得到葛根总黄酮的得率为1.47%。该实验为以后研发新的葛根及葛藤茎产品提供了理论依据。In this paper,with wild Pueraria lobata stems and Pueraria lobata roots as the raw materials,the volatile oil components of Pueraria lobata stems are extracted by steam distillation-ether extraction method,and then analyzed by gas chromatography-mass spectrometry(GC/MS).Taking the yield of total flavonoids as the index,rutin as the control sample,and sodium nitrite-aluminum nitrate method as the analysis method of flavonoids,the total flavonoids from Pueraria lobata roots are extracted by using ultrasonic-assisted extraction.The effects of ethanol concentration,solid-liquid ratio,ultrasonic power,extraction time and extraction temperature on the yield are investigated by single factor experiment,and orthogonal experiment is used to determine the extraction process of total flavonoids from Pueraria lobata roots.The results show that 10 main volatile components are successfully isolated and identified from Pueraria lobata stems,accounting for 83.94%of the total components,among which,alcohols account for 49.68%,esters account for 15.85%,alkanes account for 5.81%,and phenols account for 12.60%.Among them,the content of globulol is the highest,accounting for 31.30%.The standard curve equation of rutin sample is Y=11.146X-0.006,R^(2)=0.9992.The RSD of the precision and stability experiment of extracts from Pueraria lobata roots is 0.227%and 1.23%respectively.The average recovery rate is 98.6%,and the RSD is 1.86%.The optimal extraction process of total flavonoids from Pueraria lobata roots is extraction temperature 80℃,ethanol concentration 40%,extraction time 20 min,solid-liquid ratio 1∶50(g/mL)and ultrasonic output power 300 W.Under these conditions,the yield of total flavonoids from Pueraria lobata roots is 1.47%.This experiment has provided a theoretical basis for the future development of new products of Pueraria lobata roots and Pueraria lobata stems.
