摘  要:目的 探讨人重组Shh蛋白(SHH)信号通路对大脑中动脉梗死(MCAO)模型大鼠脑缺血后的保护机制。方法 SPF级SD大鼠36只随机分为假手术组、模型组和SHH组,每组各12只。大鼠MCAO模型制备采用线栓法制成,侧脑室注射外源性SHH蛋白激活SHH信号通路。神经功能依据Longa标准评分进行评价;采用TTC测定大鼠梗死体积;采用激光多普勒血流仪,记录左侧大脑中动脉供血区域血流变化值;实时荧光定量PCR法(RT-PCR)测定VEGF和Ang-1 mRNA表达;采用Western blot测定VEGF和Ang-1蛋白表达。结果 模型组和SHH组大鼠神经功能评分高于假手术组(P<0.05);SHH组大鼠神经功能评分低于模型组(P<0.05)。模型组和SHH组大鼠梗死体积高于假手术组(P<0.05);SHH组大鼠梗死体积低于模型组(P<0.05)。模型组和SHH组大鼠脑血流值低于假手术组(P<0.05);SHH组大鼠脑血流值高于模型组(P<0.05)。模型组和SHH组大鼠VEGF和Ang-1相对表达量低于假手术组(P<0.05);SHH组大鼠VEGF和Ang-1相对表达量高于模型组(P<0.05)。模型组和SHH组大鼠VEGF和Ang-1蛋白表达灰度值低于假手术组(P<0.05);SHH组大鼠VEGF和Ang-1蛋白表达灰度值高于模型组(P<0.05)。结论 SHH信号通路可改善MCAO大鼠神经功能,降低梗死体积,通过上调VEGF和Ang-1表达促进血管新生。Objective To investigate the protective mechanism of human recombinant Shh protein(SHH) signal pathway on middle cerebral artery infarction(MCAO) model rats after cerebral ischemia. Methods A total of 36 SPF SD rats were randomly divided into the sham operation group, the model group and the SHH group, with 12 rats in each group.The MCAO rat model was prepared using the thread plug method. The SHH signal pathway was activated by injection of exogenous SHH protein into the lateral ventricle. Neural function was evaluated according to Longa standard score.Infarct volume was measured by TTC. Laser Doppler blood flow meter was used to record blood flow changes in the blood supply area of the left middle cerebral artery. Real-time fluorescent quantitative PCR(RT-PCR) was used to measure VEGF and Ang-1 m RNA expression. Western Blot was used to determine VEGF and Ang-1 protein expression. Results The neurological function scores in the model group and the SHH group were higher than that in the sham operation group(P<0.05). The neurological function score in the SHH group was lower than that in the model group(P<0.05). The infarct volumes in the model group and the SHH group were higher than that in the sham operation group(P<0.05). The infarct volume in the SHH group was lower than that in the model group(P<0.05).The cerebral blood flow values in the model group and the SHH group were lower than that in the sham operation group(P<0.05).The cerebral blood flow value in the SHH group was higher than that in the model group(P<0.05). The relative expression levels of VEGF and Ang-1 in the model group and the SHH group were lower than those in sham operation group(P<0.05). The relative expression levels of VEGF and Ang-1 in SHH group were higher than those in the model group(P<0.05).The gray values of VEGF and Ang-1 protein expression in the model group and the SHH group were lower than that in the sham operation group(P<0.05).
