摘  要:目的研究PYCR基因家族(PYCRs)与乳腺癌发生、发展及预后的关系。方法应用Oncomine,基于基因表达水平值的交互分析平台(GEPIA)和人类蛋白图谱数据库以及荧光定量聚合酶链反应实验分析PYCRs在乳腺癌中的表达情况;通过Kaplan-Meier Plotter和肿瘤免疫在线研究工具(TIMER)数据库分析PYCRs的表达与乳腺癌临床病理分期以及预后的关系。结果PYCRs在乳腺癌中均为高表达;PYCRL与乳腺癌病理分型显著相关;PYCR1与PYCRL的高表达具有较差的预后;PYCRs与乳腺癌肿瘤纯度呈正相关,而与多种免疫细胞浸润丰度呈负相关。结论本研究发现PYCRs的高表达与乳腺癌的发生发展密切相关,该基因家族有望成为乳腺癌的诊断标志物以及免疫治疗的关键靶点。Objective To investigate the relationship between pyrrolinre-5-carboxylate reductases(PYCR)gene family and the occurrence,development and prognosis of breast cancer.Methods Oncimie,GEPIA and HPA databases and fluorescence quantitative PCR assay were used to analyze the expression of PYCRs in breast cancer.Kaplan-Meier Plotter and TIMER databases were used to analyze the relationship between the expression of PYCRs and the clinical pathological staging and prognosis of breast cancer.Results PYCRs were highly expressed in breast cancer.PYCRL was significantly related to the pathological classification of breast cancer.The high expression of PYCR1 and PYCRL showed poor prognosis.PYCRs were positively correlated with the purity of breast cancer tumors,and negatively correlated with infiltrating abundance of various immune cells.Conclusion This study finds that the high expression of PYCRs is closely related to the occurrence and development of breast cancer,and the gene family is promising as a diagnostic marker for breast cancer and a key target for immunotherapy.
