摘  要:目的分析非小细胞癌组织中β-连环蛋白(β-catenin)和程序性死亡配体1(PD-L1)在mRNA和蛋白水平中的表达,探讨二者表达水平之间的关系。方法用qRT-PCR方法检测非小细胞肺癌组织和配对的癌旁组织中β-catenin和PD-L1 mRNA表达水平;用免疫组织化学方法检测β-catenin和PD-L1蛋白表达水平,通过染色强度来评价其蛋白水平。结果和配对的癌旁组织比较,非小细胞肺癌组织中β-catenin(18/35)和PD-L1(20/35)mRNA表达水平明显升高,二者之间呈正相关(r=0.582);非小细胞肺癌组织中β-catenin(17/35)和PD-L1(22/35)蛋白表达均升高,且二者的免疫组化染色强度一致。结论非小细胞肺癌组织中β-catenin和PD-L1在m RNA和蛋白水平中表达明显升高,二者之间呈正相关,Wnt/β-catenin信号通路可能参与了非小细胞肺癌中PD-L1的表达调控,促进肿瘤进展。Objective The aim of this study is to determine the expression levels of the mRNA and protein of β-catenin and PDL1 genes in non-small cell lung cancer(NSCLC)tissues and paired adjacent non-cancer tissues and explore the relationship between the expression levels of β-catenin and PD-L1 genes.Methods The expression levels of β-catenin and PD-L1 mRNA in non-small cell lung cancer tissues and paired adjacent tissues were detected by qRT-PCR.The protein expression levels of β-catenin and PD-L1 were detected by immunohistochemistry,and the protein levels were evaluated by staining intensity.Results The mRNA expression levels of β-catenin(18/35)and PD-L1(20/35)in NSCLC tissues were significantly higher than those in adjacent tissues,and there was a positive correlation between them(r=0.582).The expression of β-catenin(17/35)and PD-L1(22/35)protein were increased in NSCLC tissues,and their immunohistochemical staining intensity was consistent.Conclusion The mRNA and protein levels of β-catenin and PD-L1 were significantly increased in NSCLC tissues,and there was a positive correlation between the two.The Wnt/β-catenin signaling pathway may be involved in the regulation of PD-L1 expression in NSCLC and promote tumor progression.
