摘 要:目的建立基于激光捕获显微切割(laser capture microdissection,LCM)-Agilent miRNA芯片技术的结肠癌化疗耐药microRNA(miRNA)表达谱,筛选结肠癌化疗耐药相关靶标。方法应用LCM技术对Ⅲ期结肠癌化疗后复发/未复发的肿瘤实质细胞进行细胞纯化。Agilent miRNA芯片检测两种细胞中的miRNA表达情况,根据差异表达倍数和功能预测筛选结肠癌化疗相关靶标。结果包括miR-194-3p、miR-3607、miR-887、miR-424-5p、miR-1237等16个miRNA为差异基因,可作为潜在结肠癌化疗耐药相关靶标进一步研究。q RT-PCR证实,miR-194-3p在结肠癌化疗后复发样本中高表达(P<0.001)。结论运用LCM结合Agilent miRNA芯片技术筛选出16个与结肠癌化疗耐药相关的miRNA,且miR-194-3p在结肠癌化疗后复发样本中高表达,是潜在结肠癌化疗耐药治疗靶标。Objective This study aims to establish the chemotherapy resistance microRNA(miRNA)expression profile for colon cancer patients based on laser capture microdissection and Agilent miRNA chip technology and screen chemotherapy resistance related target.Methods Laser capture microdissection were used for tumor cells purification in stage Ⅲ recurrent and no recurrent colon cnacer patients after chemotherapy.Furthermore,the expression patterns and gene function prediction for target miRNA were analyzed.Results A total of 16miRNA,including miR-194-3p,miR-3607,miR-887,miR-424-5p,miR-1237 were identified as differentially expressed genes,which might be novel chemotherapy resistance related target in colon cancer.Results of q RT-PCR indicated that miR-194-3p was up-regulated in carcinoma recurrence patients after chemotherapy(P<0.001).Conclusion We screened 16 chemotherapy resistance related miRNA for colon cancer which based on laser capture microdissection and Agilent miRNA chip technology.Furthermore,we identified miR-194-3p expression patterns in recurrence patients after chemotherapy for colon cancer,which represents potential therapy target for chemoresistance.
