抗体特异性抑制赖氨酰氧化酶对胃癌SGC-7901转移能力的影响

摘　　要：目的探讨抗体特异性抑制赖氨酰氧化酶(lysyl oxidase,LOX)作用于胃癌SGC-7901细胞后,基质金属蛋白酶-9(matrix metalloproteinases-9,MMP-9)的变化和体外黏附、迁移的改变。方法培养胃癌SGC-7901细胞,用0、5、10和15μg·mL^(-1)的LOX特异性抗体(anti-LOX)干预细胞,Western blot法检测MMP-9的相对表达量,同质黏附实验和异质黏附实验检测细胞黏附能力,体外划痕实验检测细胞迁移能力。结果15μg·mL^(-1) anti-LOX干预后,胃癌细胞MMP-9的表达降低(P<0.05);0、5、10和15μg·mL^(-1)的anti-LOX干预后,细胞的同质黏附数量随着anti-LOX浓度的升高而增多(P均<0.05),异质黏附数量随着anti-LOX浓度的升高而减少(P均<0.05);anti-LOX干预后,各个时间点SGC-7901迁移细胞个数随抗体浓度的升高而减少(P均<0.05)。结论抗体特异性抑制LOX作用于胃癌细胞后,可增强同质黏附、减弱异质黏附、下调体外迁移能力。Objective To study the effects of antibody specific inhibition of lysyl oxidase(LOX)on matrix metalloproteinase-9(MMP-9)production,adhesion and migration of gastric cancer cell line SGC-7901.Methods Gastric cancer cell line SGC-7901 was cultured and treated with 0μg·mL^(-1),5μg·mL^(-1),10μg·mL^(-1),15μg·mL^(-1) LOX-specific antibody(anti-LOX).The production of MMP-9 was checked by Western blot.The adhesion of SGC-7901 was analyzed by homogeneous adhesion test and heterogeneous adhesion test.The migration of SGC-7901 was measured by scratch test in vitro.Results After treated with 15μg·mL^(-1) anti-LOX,the expression of MMP-9 in gastric cancer cells decreased(P<0.05).When SGC-7901 were treated with 0μg·mL^(-1),5μg·mL^(-1),10μg·mL^(-1),15μg·mL^(-1) anti-LOX,the cell number of homogeneous adhesion gradually increased with the augment of anti-LOX(P all<0.05),and the cell number of heterogeneous adhesion decreased with the augment of anti-LOX(P all<0.05),the cell number of migratory decreased with the augment of antibody at each time point(P all<0.05).Conclusion Suppression of LOX using specific antibody may inhibit tumor metastasis by enhancing homogeneous adhesion,weakening heterogeneous adhesion and attenuate migration in vitro.
