摘 要:目的:探讨高迁移率族蛋白A1(high mobility group AT-hook 1,HMGA1)和微小RNA(microRNA,miRNA)let-7i-5p对三阴性乳腺癌细胞的影响及可能的分子机制。方法:运用在线生物信息学工具GEPIA分析HMGA1在乳腺癌组织中的表达情况。选择人三阴性乳腺癌MDA-MB-231细胞,将其分为阴性对照组(negative control,NC)、siRNA HMGA1(si-HMGA1)组和si-HMGA1+let-7i-5p抑制物(si-HMGA1+let-7i-5p inhibitor)组;采用qRT-PCR法检测转染后let-7i-5p、HMGA1的mRNA水平;采用Western blot法检测转染后HMGA1的蛋白表达水平;CCK-8法检测转染后各组细胞的增殖活力;划痕实验检测转染后各组细胞的划痕愈合率;Transwell实验检测转染后各组细胞的侵袭数量。结果:数据库证实HMGA1在乳腺癌组织中的表达量显著高于正常组织(P<0.05)。MDA-MB-231细胞转染后,与NC组比较,si-HMGA1组细胞中let-7i-5p水平显著升高,HMGA1的mRNA和蛋白水平显著降低;细胞的增殖活力、划痕愈合率和侵袭细胞的数量均显著降低(P<0.05)。与si-HMGA1组比较,si-HMGA1+let-7i-5p inhibitor组细胞的增殖、迁移和侵袭的能力显著增加(P<0.05)。结论:HMGA1低表达可抑制三阴性乳腺癌细胞的增殖和侵袭,且let-7i-5p可调控HMGA1对三阴性乳腺癌细胞的影响。Objective:To investigate the effects of high mobility group AT-hook 1(HMGA1)and microRNA(miRNA)let-7i-5p on triple-negative breast cancer cells and their possible molecular mechanisms.Methods:The expression of HMGA1 in breast cancer tissues was analyzed by online bioinformatics tool GEPIA.The triple negative breast cancer MDA-MB-231 cells were selected and divided into negative control(NC)group,siRNA HMGA1(si-HMGA1)group,and si-HMGA1+let-7i-5p inhibitor group respectively.qRT-PCR was used to detect the levels of let-7i-5p and HMGA1 mRNA after transfection.The protein expression level of HMGA1 after transfection was detected by Western blot.CCK-8 method was used to detect the proliferation activity of transfected cells.The scratch healing rate of transfected cells was detected by scratch test.Transwell test was used to detect the number of invasion cells in each group after transfection.Results:The database confirmed that the expression of HMGA1 in breast cancer tissues was significantly higher than that in normal tissues(P<0.05).After MDA-MB-231 cells were transfected,compared with the NC group,the level of let-7i-5p in si-HMGA1 group increased significantly,and the mRNA and protein levels of HMGA1 decreased significantly.And the cell proliferation activity,scratch healing rate and the number of invasive cells also decreased significantly(P<0.05).Compared with si-HMGA1 group,the proliferation,migration and invasion of cells in si-HMGA1+let-7i-5p inhibitor group significantly increased(P<0.05).Conclusion:The down-regulation of HMGA1 expression can inhibit the proliferation and invasion of triple-negative breast cancer cells,and let-7i-5p can regulate the effect of HMGA1 on triple-negative breast cancer cells.
