摘　　要：目的探讨蛋白精氨酸甲基转移酶5(PRMT5)调控非小细胞肺癌(NSCLC)细胞增殖、生长的作用,旨在为NSCLC的诊断与治疗提供新思路。方法利用慢病毒感染的方法构建PRMT5过表达稳定转染A549细胞系,通过MTT和平板克隆实验探讨PRMT5对A549增殖、生长的影响,并分析PRMT5小分子抑制剂C9抑制PRMT5表达后对A549增殖、生长的影响。结果利用PRMT5慢病毒感染A549细胞,Western blot和qRT-PCR实验结果显示,PRMT5蛋白、mRNA表达显著上调。过表达PRMT5可提高A549的增殖、生长能力。抑制PRMT5表达可降低A549的增殖、生长能力。结论PRMT5参与调控NSCLC细胞的增殖、生长,可能成为诊断NSCLC的新型肿瘤标志物及阻断或延缓NSCLC进展的潜在治疗靶点。Objective To investigate the role of protein arginine methyltransferase 5(PRMT5)in regulating the proliferation and growth of non-small cell lung cancer(NSCLC)cells,in order to provide new ideas for the diagnosis and treatment of NSCLC.Methods PRMT5 overexpression stably transfected A549 cell line was constructed by lentivirus infection method.The effects of PRMT5 on the proliferation and growth of A549 were explored by MTT and plate cloning experiments,the effects of PRMT5 small molecule inhibitor C9 inhibiting the expression of PRMT5 on the proliferation and growth of A549 were analyzed.Results PRMT5 lentivirus was used to infect A549 cell,and Western blot and qRT-PCR experimental results showed that the expression of PRMT5 protein and mRNA significantly up-regulated.Overexpression of PRMT5 could improve the proliferation and growth ability of A549.Inhibition of PRMT5 expression could inhibit the proliferation and growth ability of A549.Conclusion PRMT5 is involved in regulating the proliferation and growth of NSCLC cells,and may be a new tumor marker for the diagnosis of NSCLC and a potential therapeutic target for blocking or delaying the progression of NSCLC.
