耐药性嗜水气单胞菌感染致四眼斑龟死亡的病例报告

摘　　要：某养殖户所饲四眼斑龟(Sacalia quadriocellata)突发群体死亡,为查明具体死亡原因,提供有效的后续防控措施建议,本试验对病死的四眼斑龟进行大体剖检,取相应脏器制成病理切片进行观察,于多脏器中获取病料进行细菌分离培养,经PCR扩增、全基因组测序、药敏试验和动物回归感染试验,确定致病病原体。结果显示:引起四眼斑龟发病的病原体为1株耐氨苄青霉素的嗜水气单胞菌(Aeromonas hydrophila),其对氨苄青霉素和喹乙醇具有耐药性;分离菌的基因组中含有2种与β-内酰胺酶相关的耐药基因和多种毒力基因;在动物回归感染试验中,接种该株嗜水气单胞菌的小鼠出现了被毛粗乱、食欲减退,部分小鼠死亡;经计算得该分离菌的半数致死量为2.00×10^(7) CFU,主要侵袭小鼠的肝脏、肾脏和肺脏。据此判断,该起四眼斑龟发病事件由分离所得的耐药性嗜水气单胞菌感染引起。In order to determine the cause of sudden death of four-eyed turtles(Sacalia quadriocellata) from a farm and to advice on follow-up prevention and control measures,this study conducted an autopsy of dead turtles,carried out pathological examination of tissue sections from corresponding organs,and performed bacterial isolation from multiple organs followed by pathogen identification through PCR,whole genome sequencing analysis,drug sensitivity test and animal regression infection test.The results showed that a Gram-negative bacterium was isolated and confirmed as Aeromonas hydrophila,which demonstrated resistance to ampicillin and olaquindox;the isolate harbored two antibiotic-resistant genes related to β-lactamase and several other virulence genes in the genome;in the animal regression infection test,the mice inoculated with the Aeromonas hydrophila showed coarse hair,decreased appetite,and some mice died;the median lethal dose of the isolate was 2.00×10^(7) CFU,which mainly invaded the liver,kidney and lung of mice.Thus,the death of Sacalia quadriocellata was likely caused by ampicillin and olaquindox-resistant A.hydrophila.
