摘　　要：目的:探究纺锤体装配检查点蛋白家族成员BUB1B在肺鳞状细胞癌(LUSC)中的表达及其意义。方法:收集基因表达综合数据库(GEO)、癌症基因组图谱(TCGA)数据库中BUB1B在LUSC中的表达值,通过计算标准化均数差(SMD)及绘制综合受试者工作特征曲线(sROC),综合分析BUB1B mRNA在公共数据集中的表达情况,用实时荧光定量聚合酶链式反应(RTqPCR)验证BUB1B在LUSC细胞系(NCI-H2170)与人正常肺上皮细胞系(BEAS-2B)中BUB1B mRNA的表达,免疫组化技术(IHC)验证61例LUSC组织和对应54例癌旁/正常肺组织中BUB1B蛋白的表达情况,搜索癌症中的增强子RNA(eRic)数据库在LUSC中与BUB1B表达相关增强子RNA(eRNA),RT-qPCR验证eRNA在NCI-H2170和BEAS-2B中mRNA的表达。结果:BUB1B mRNA在公共数据集中均呈现出在LUSC组织中的高表达趋势(P<0.05)。BUB1B在LUSC细胞系中的表达高于人正常肺上皮细胞系(P<0.05)。BUB1B蛋白在LUSC组织中高表达(P<0.05),且在原发肿瘤大小、临床分期中的差异表达具有统计学意义(P<0.05),在性别、年龄、淋巴结转移、远处转移、分化程度、吸烟史中的差异表达无统计学意义(P>0.05)。ENSR00000075512在LUSC组织中与BUB1B表达相关,且ENSR00000075512 RNA在LUSC细胞系中的表达高于人正常肺上皮细胞系(P<0.05)。结论:BUB1B在LUSC中高表达,可能通过与ENSR00000075512相互作用参与LUSC的发生发展。Objective:To investigate the expression of BUB1B,a member of the spindle assembly checkpoint protein family,in lung squamous cell carcinoma(LUSC)and its clinical significance.Methods:The expression values of BUB1B in LUSC were collected from the Gene Expression Omnibus(GEO)and The Cancer Genome Atlas(TCGA)databases,and the expression of BUB1B mRNA in the public dataset was analyzed comprehensively by calculating the standardized mean difference(SMD)and plotting the summary receiver operating characteristic(SROC)curve.Real-time quantitative PCR(RT-qPCR)was used to validate BUB1B mRNA expression in LUSC cell line(NCI-H2170)versus human normal lung epithelial cell line(BEAS-2B).Immunohistochemistry(IHC)to validate BUB1B protein expression in 61 LUSC tissues and corresponding 54 paraneoplastic/normal lung tissues.The enhancer RNA(eRNA)in cancer(eRic)database was used to search for eRNA associated with BUB1B expression in LUSC,and RT-qPCR was conducted to verify the mRNA expression of eRNA in NCIH2170 and BEAS-2B.Results:BUB1B mRNA showed a trend of high expression in LUSC tissues in both public datasets(P<0.05).The expression of BUB1B was higher in LUSC cell lines than that in human normal lung epithelial cell lines(P<0.05).The BUB1B protein was highly expressed in LUSC tissues(P<0.05),and the differential expression with varying primary tumor size and clinical stage was statistically significant(P<0.05),while the differences in expression with varying gender,age,lymph node metastasis,distant metastasis,degree of differentiation,and smoking history were not statistically significant(P>0.05).ENSR00000075512 was correlated with BUB1B expression in LUSC tissues,and the expression of ENSR00000075512 RNA in LUSC cell lines was higher than that in human normal lung epithelial cell lines(P<0.05).Conclusion:BUB1B is highly expressed in LUSC and may be involved in the development of LUSC by interacting with ENSR00000075512.
