摘  要:对内生拮抗细菌264ZY7的生长温度、pH值、生长周期、固氮、溶磷及产IAA能力等生物学特性进行测定,并结合单因素试验和正交设计试验对其发酵条件进行优化。结果表明,菌株264ZY7具有稳定的固氮和产IAA能力;在NA培养基上菌株264ZY7最高生长温度55℃、最低温度2℃、致死温度110℃和最适温度30~36℃,最适pH 5~7,在5 h进入对数生长期,在10~14 h时进入稳定生长期;其最优发酵条件为:牛肉膏3 g、蔗糖25 g、NaCl 5 g、酵母膏5 g、水1000 mL;温度28℃,接种量1%,装液量40 mL·150 mL^(-1),摇床转速240 r·min^(-1),pH 6,100 L发酵罐最佳放罐时间为21 h。获得了最佳培养条件,该结果为利用菌株264ZY7开发生防制剂奠定了基础。The optimal culture method of strain 264ZY7 was established through measuring the biological characteristics of the strain and optimizing the fermentation.Biological features of strain 264ZY7,such as growth and reproduction temperature,pH,growth cycle,nitrogen fixation,phosphorus solubilization,IAA-producing ability,were determined and fermentation conditions were optimized by single factor and orthogonal test.The results showed that strain 264ZY7 on NA medium had stable nitrogen fixation and IAA production capacity,its maximum growth temperature was 55℃and a minimum temperature was 2℃,the lethal temperature was 110℃,and the optimum temperature were 30-36℃.The optimum pH was 5-7.The growth curve of strain 264ZY7 showed that it entered the logarithmic growth phase at 5 h and the stable growth phase at 10-14 h.The optimal fermentation conditions of 264ZY7 were 3 g beef grit,25 g sucrose,5 g NaCl,5 g yeast extract,1000 mL water,28℃temperature,1%inoculum volume,40 mL·150 mL^(-1) liquid volume,240 r·min^(-1) shaker speed,pH 6,and the optimal released time for the 100 L fermenter tank is 21 h.The culture conditions of strain 264ZY7 were optimized,which laid the foundation for the development of anti-formulation.
