摘 要:目的:研究卵巢癌SKOV3细胞及其铂类耐药SKOV3/DDP细胞中整合素α5(integrinα5,ITGA5)的表达情况,探索ITGA5沉默后对卵巢癌SKOV3/DDP细胞铂类敏感性、增殖、侵袭及凋亡的影响。方法:使用qRT-PCR检测卵巢癌SKOV3细胞及铂类耐药细胞SKOV3/DDP中的ITGA5表达情况。随后将SKOV3/DDP细胞分别分为siITGA5-SKOV3/DDP组(ITGA5沉默组)、siNC组(阴性对照组)和对照组,siITGA5-SKOV3/DDP组和siNC组分别转染ITGA5沉默质粒si ITGA5和阴性对照质粒siNC,对照组给予空载体。采用CCK8法检测各组细胞顺铂的半数致死率(50%inhibitory concentration,IC50)、细胞增殖活性;Transwell实验检测各组细胞侵袭能力;流式细胞术检测各组细胞的凋亡情况。结果:卵巢癌铂类耐药细胞SKOV3/DDP中ITGA5表达明显高于SKOV3细胞,差异有统计学意义(P<0.05);siITGA5-SKOV3/DDP组、siNC组及对照组顺铂的IC50分别为2.096μg/mL、2.507μg/mL及2.619μg/mL,差异有统计学意义(P<0.05);与siNC组及对照组细胞系相比,siITGA5-SKOV3/DDP组细胞第5、6、7天的细胞增殖活性明显下降,同时侵袭能力亦明显下降,细胞凋亡率上升,差异有统计学意义(P<0.05)。结论:ITGA5在人卵巢上皮性癌铂类耐药细胞SKOV3/DDP中表达升高,ITGA5表达下降后SKOV3/DDP对顺铂的敏感性增加,增殖和侵袭能力下降;ITGA5可能通过抑制卵巢癌SKOV3/DDP细胞凋亡参与铂类的耐药。Objective:To investigate the expression of integrated alpha 5(ITGA5) in ovarian cancer SKOV3 cells and platinum-resistant SKOV3/DDP cells,in order to explore the effects of ITGA5 on platinum-sensitivity,proliferation,invasion and apoptosis of SKOV3/DDP cells.Methods:qRT-PCR was performed to investigate the expression level of ITGA5.SKOV3/DDP cells were assigned to the siITGA5-SKOV3/DDP group(ITGA5-silenced),the siNC group(negative control group) and the control group,respectively.siITGA5-SKOV3/DDP group and siNC group were transfected with ITGA5-silenced plasmids(siITGA5) and negative control plasmids,and the control group was injected with empty vector.Cell proliferation and 50% inhibitory concentration(IC50) of cisplatin were detected by CCK8 assays,invasion by Transwell assays,and apoptosis by flow cytometry analysis.Results:The expression level of ITGA5 in SKOV3/DDP cells were higher than that in SKOV3 cells(P<0.05).The IC50 of cisplatin in the siITGA5-SKOV3/DDP group,the siNC group and the control group were 2.096 μg/mL,2.507 μg/mL and 2.619 μg/mL,respectively,and the difference among groups was statistically significant(P<0.05).Compared with the siNC group and the control group,proliferation and invasion of siITGA5-SKOV3/DDP cells significantly decreased at 5 th,6 th and 7 th days(P<0.05),while the apoptosis rate increased(P<0.05).Conclusion:The expression of ITGA5 in ovarian cancer SKOV3/DDP cells increases,and the sensitivity to cisplatin,proliferation and invasion of SKOV3/DDP cells will decrease by downregulating the expression of ITGA5.ITGA5 may be involved in platinum resistance by inhibiting the apoptosis of ovarian cancer SKOV3/DDP cells.
